Functional and transcriptional profiling of MUTZ‐3, a myeloid cell line acting as a model for dendritic cells

Abstract: T cells, whereas KG-1 and THP-1 only induced a marginal response. Furthermore, MUTZ-3 displayed the phenotypic and transcriptional profiles of immature dendritic cells, after differentiation with granulocyte-macrophage colony-stimulating factor and interleukin-4. Upon activation with inflammatory cytokines, MUTZ-3 matured phenotypically and exhibited a gene induction similar to that of monocyte-derived dendritic cells. This delineation of the cellular and transcriptional activity of MUTZ-3, in response to matu… Show more

“…In the current research, the myelomonocytic cell line, MUTZ-3, was assessed for the ability to respond to sensitizing agents and the expression of biomarkers investigated. When compared to other DC-like cell lines used in current research, such as THP-1 cells, MUTZ-3 represent the most physiologically similar cells to in vivo DCs (Larsson et al, 2006).…”

“…To avoid such issues, human cell lines, with characteristics similar to DCs, such as THP-1 and MUTZ-3 cells have been utilised (Sakaguchi et al, 2009). MUTZ-3 comprises the most physiologically similar cell line to its in vivo DC counterpart and represents the immortalised equivalent of CD34 + DC precursors (Larsson et al, 2006). Current assay development utilises a test panel of chemicals consisting of extreme, strong, intermediate and weak sensitizers with non-sensitizers all of which have been classified by the local lymph node assay (LLNA) in order to correlate in vitro data with known in vivo data.…”

“…5,6 The K562 (chronic myelogenous leukemia) and THP-1 (monocytic leukemia) cell lines were cultured in Roswell Park Memorial Institute 1640 medium supplemented with 10% fetal bovine serum, 25 mM 4-(2-hydroxyethyl)-1-piperazine ethanesulfonic acid, and 2 mM glutamine (complete medium), at 37°C in a humidified atmosphere with 5% CO 2 . MUTZ-3 cells [23][24][25] were maintained in complete medium supplemented with granulocyte-macrophage colony-stimulating factor (10 ng/mL) and their dendritic differentiation was induced in the presence of 1,000 U/mL interleukin-4 (IL-4) for 5 days.…”

“…To generate immature MoDC, human peripheral blood monocytes were further cultured for 7 days in RPMI 1640 medium containing 10% FCS (PAA laboratories, Linz, Austria), 2% human serum, 150 ng/ml rGM-CSF and 50 ng/ml IL-4 (PeproTech EC, London, UK). Half of the medium volume was exchanged every 2-3 days [10].…”
