The structures were determined by spectroscopic methods, mainly 1D and 2D NMR.The rhizomes of Polygonatum cyrtonema Hua, one of the original plants named "Huang-jing" in traditional Chinese medicine, are used as a tonic remedy to treat lung and stomach ailments. Steroidal saponins with structurally diverse and remarkable biological activities have attracted much attention [1,2], and a number of them have been isolated from other original plants of "Huang-jing" [3,4], but no chemical studies have been performed on P. cyrtonema. To discover biologically and chemically novel agents, we chose the rhizomes of P. cyrtonema for phytochemical investigation.In this study, the ethanol extract of the rhizomes led to the isolation of three pairs of mixture of 25R and 25S stereoisomeric spirostanol saponins: (25R) spirostan-5-en-12-(2a, 2b) [6,7], and (25R, 25S)-3-E-hydroxyspirost-5-en-12-one (3a, 3b) [6,8]. Compound 1a is a new compound. Compound 2b was first found from the Polygonatum species, and compounds 1b, 2a, 3a, and 3b were found for the first time from this plant.
