Genetic characterisation and population structure analysis of Anatolian figs (Ficus carica L.) by SSR markers

Abstract: The common fig (Ficus carica L.) is a tree species and is one of the oldest fruit trees cultivated in Turkey. The Western Anatolian region of Turkey produces nearly a quarter of the total dried fig production of the world. This region also harbours a rich fig germplasm. However, so far this germplasm has remained largely uncharacterised. In this study, using 14 simple sequence repeat (SSR) primer pairs, we analysed a total of 310 fig accessions from six different regions of Anatolia. In structure analyses, Wes… Show more

“…There does not appear to be any consensus for a common list of SSRs that can be used in fig genotyping. Consequently, we compared 10 papers on fingerprinting of fig collections (Akin et al., 2021; Baraket et al., 2011; Boudchicha et al., 2018; Ergül et al., 2021; Ferrara et al., 2016; Knap et al., 2018; Marcotuli et al., 2019; Perez‐Jiménez et al., 2012; Poljiula et al., 2021; Rodolfi et al., 2018) and ranked 22 SSRs according to the frequency with which they had been adopted by different authors (Table 16). The top marker among those listed was the one that was most commonly used (8 out of 10 authors) and others were then ranked in descending order.…”

“…DNA was extracted from leaf tissue as described by Lefort et al [22]. Determination of DNA concentration and quality was performed according to Akçay et al [23] and Ergül et al [24] using an ND-1000 spectrophotometer (NanoDrop Technologies, Wilmington, DE, USA) and agarose (1%, w/v) gel electrophoresis. Isolated DNA was stored at −20 • C until PCR reactions were performed.…”

“…We also used STRUCTURE-HARVESTER [43] to estimate the best K value supported by the current data [44]. Similar to Ergül et al [24], the number of accessions strongly assigned to each RPP was determined based on the qI probabilities (probability of membership) higher than 80%.…”
