Genetic diversity and population structure of Butea monosperma (Lam.) Taub.- a potential medicinal legume tree

Abstract: Three molecular marker systems, Random Amplified Polymorphic DNA (RAPD), Inter-Simple Sequence Repeats (ISSR) and Sequence-Related Amplified Polymorphism (SRAP) were employed to investigate the genetic structure and diversity among the 14 natural populations of Butea monosperma collected from different geographical regions of India. Detected by 17 RAPD, 15 ISSR and 11 SRAP primer combinations, the proportions of polymorphic bands were 84.2 %, 77.2 % and 91.9 %, respectively, and the mean Nei's genetic distance… Show more

“…The structure of Moroccan populations, calculated by AMOVA, had revealed a high level of variation within populations (85%) and a low level of variation among populations (11%). Our results are in line with the previous study on genetic diversity of T. articulata (Sánchez-Gómez et al, 2013) and other Cupressaceae species (Meloni et al, 2006;Liu et al, 2013;Hou et al, 2018) that maintain more variation within species and within populations species than species with other life forms but have less variation among populations (Vashishtha et al, 2013). The genetic differentiation values detected for gymnosperms (Gst = 0.18) (Nybom and Bartish, 2000) are similar to our Genetic structure of 129 genotypes assessed by STRUCTURE software (20 independent runs of each K value (from K = 1 to 10) were performed with 50,000 as the burn-in and 100,000 iterations.…”

“…For instance, Zhang et al (2012) estimated 96 polymorphic bands for Ormosia hosiei, in Sapindus trifoliatus 185 bands were polymorphic (Mahar et al, 2013), and in Butea monosperma 71 bands were generated (Vashishtha et al, 2013). Therefore, a bootstrap analysis has been used in order to verify whether this number of polymorphic bands is sufficiently able to respond precisely to the estimative obtained in each study.…”

“…Currently there are several techniques available that allow the study of genetic diversity in plants, including morphological, biochemical and molecular markers (Vashishtha et al, 2013). The most successful of those that have been developed and put to use in the last two decades are molecular markers based on the polymerase chain reaction technique (PCR), such as Simple Sequence Repeat (SSR), Random Amplified Polymorphic DNA (RAPD) and Inter-Simple Sequence Repeat (ISSR) (Semagn et al, 2006).…”
