Genetic homogeneity revealed in micropropagated Bauhinia racemosa Lam. using gene targeted markers CBDP and SCoT

Abstract: Two gene targeted markers i.e. CAAT boxderived polymorphism (CBDP) and start codon targeted (SCoT) polymorphism were applied to analyze the genetic stability of in vitro propagated plants of Bauhinia racemosa Lam. multiplied by enhanced axillary shoot proliferation of mature tree derived nodal explant. Nine randomly selected micropropagated plants of 1 year age were subjected to molecular analysis. The isolated genomic DNA samples were subjected to PCR amplification with a total of 61 primers (25 CBDP and 36 S… Show more

“…In a previous study, high degree of genetic variation in plantlets of Rumex nepalensis, micropropagated via direct and indirect shoot organogenesis was detected by 12 SCoT primers, whereas it was undetected using 7 RAPD markers (Bhattacharyya et al, 2017a). Genetic stability in 9 randomly propagated plants of Bauhinia racemosa using 18 SCoT makers was reported as true-to-type (Sharma et al, 2019).…”

“…SCoT (start codon targeted marker) is a novel technique to confirm homogeneity of micropropagated plantlets derived from organogenesis or embryogenesis. In addition, two or three molecular markers, confirming genetic stability, have been well established in many plant species (Bhattacharyya et al, 2017a,b;Sharma et al, 2019). The aim of present study was to induce somatic embryos from inner integuments of para rubber cv.…”

“…However, it should be kept in mind that it can stimulate genetic variability (Us-Camas et al, 2014), and thus, plants obtained through micropropagation should be carefully evaluated. Presently, this evaluation involves an array of molecular techniques (Moharana et al, 2018;Nalousi et al, 2019;Sharma et al, 2019); however, a complete comparison should also include ploidy assessment and qualitative features (Jędrzejczyk et al, 2018;Nalousi et al, 2019;Żabicki et al, 2019). Molecularly confirmed genetic fidelity only concerns fragments of the genome, not necessarily associated with trait inheritance.…”

“…Effects of environmental conditions during seed production on seed dormancy and germination have already been described. Low temperature before seed maturation may interfere with ABA biosynthesis or signaling and can markedly increase the final depth of seed dormancy (Chahtane et al, 2017;Chen et al, 2014;Kar et al, 2019;Sharma et al, 2019;Trejgell et al, 2012;Żabicki et al, 2019). However, numerous studies have shown that seeds maturing at higher temperatures produce more impermeable seeds when compared with those growing in a low-temperature environment (Jaganathan, 2016).…”

“…Genetic homogeneity of regenerated plantlets was successfully tested using Random Amplified Polymorphic DNA (RAPD) and Inter-Simple Sequence Repeat (ISSR). Recent developments in the field of plant molecular analysis have gained considerable interest in using gene targeted markers like Start Codon Targeted (SCoT) and CAAT Box Derived Polymorphism (CBDP) (Asthana et al, 2011;Rai et al, 2012;Sharma et al, 2019). RAPD and ISSR techniques depend on uncoding regions of genome whereas SCoT which is a reliable and simple dominant marker is correlated to functional genes and their corresponding traits as well (Xiong et al, 2011).…”
