Genetic Linkage Maps of Betula platyphylla Suk Based on ISSR and AFLP Markers

Abstract: Two separate genetic linkage maps for Chinese silver birch based on inter-simple sequence repeat (ISSR) and amplified fragment-length polymorphism (AFLP) were constructed by a pseudo-testcross mapping strategy. Eighty F 1 progenies were obtained from the cross between two parental trees with desirable traits (the paternal one selected from 'Qinghai' and the maternal one from 'Wangqing'). A total of 46 ISSR primers and 31 AFLP primers were employed to generate 102 ISSR and 355 AFLP polymorphic markers in the F … Show more

“…B. platyphylla is widely used not only in industry (Zhang et al 2011) but also as a medicine (Huh et al 2009). Silver birch trees are large and long-lived trees with a juvenile period of between 5 and 15 years (Wei et al 2010). However, the intensive breeding protocol has greatly reduced the length of the juvenile phase to only 2-3 years (Yang et al 2004).…”

“…Inter-simple sequence repeats (ISSRs) described by Zietkievics et al (1994) and Kantety et al (1995) have been used in genomic fingerprinting (Lüdtke et al 2010;Vargas-Ponce et al 2011), studies of genetic diversity and phylogenetic analyses (Arif et al 2009;Gomes et al 2009;Tanya et al 2011;Wei et al 2010), evolutionary biology (Wolfe et al 1998), and in several other fields. In the genus Nymphaea ISSR markers have only been used by Woods et al (2005) to study the variability and systematics of N. odorata.…”

“…The technique is simple to use providing generally reliable products (Bornet and Branchard 2001), and yield a large number of polymorphism per primer (Ye et al 2005). The products segregate mostly as dominant (Wei et al 2010) markers, but with a longer anchoring in the 5′-end some co-dominant bands have also been reported by Fisher et al (1996) and Provan et al (1996).…”

“…Under normal field conditions, birch has the highest photosynthetic capacity and growth rate among the deciduous broadleaved tree species. As a pioneer species, birch has the ability to adapt to soils with both low water content (dry and subhumid) and poor nutrient conditions (Huh et al 2009;Wei et al 2010). Since B. platyphylla is a widespread species and has many applications not only in the pulp and paper industry (Wang et al 2010b) but also as a medicine because of the antioxidant and anticancer activity of its bark extract (Ju et al 2004), it is necessary to study its development and physiology.…”
