GFP reporter gene confirms paternity in the androgenote Buenos Aires tetra,Hemigrammus caudovittatus

Abstract: A protocol for successful induction of androgenetic cloning of the Buenos Aires tetra (BT), Hemigrammus caudovittatus, with contrasting gray and golden strains is described. At the intensity of 4.2 W/m(2), UV irradiation for 2.75 min totally inactivated the maternal genome in eggs of gray BT. Following activation by sperm of golden BT, the 25-min-old embryos were shocked at 41 degrees C for 2 min to restore diploidy. Interestingly, the hatching success of the haploid fry was always higher than that of the dipl… Show more

“…Previously, a Y ‐chromosome‐specific DMRT‐1 marker has been shown to confirm the true male genotype in the unexpected female progenies sired by androgenetic males ( Y 2 Y 2 ) crossed with either androgenetic female ( X 2 X 2 ) or normal females ( X 1 X 2 ) of both BT and WT (David & Pandian, 2005 a ; unpubl. data).…”

“…Since the phenotypic characters and developmental events of BT and WT are known (David & Pandian, 2005 a ; unpubl. data), the development of hybrid embryos was monitored under stereozoom microscope (Nikon, Tokyo, Japan), especially for pigmentation of fins and body.…”

“…It utilizes single, arbitrary, decamer DNA oligonucleotide primers to amplify regions of genome based on the polymerase chain reaction (Hadrys et al 1992;Williams et al, 1993). The characters assessed through RAPD are useful for genetic studies because they provide various types of data-taxonomic population, inheritance pattern of various organisms including fishes (Brown and Epifanio, 2003;Degani, 2004;David and Pandian, 2006).…”

“…RAPD has also been used to estimate genetic diversity and variations needed to study fish management and conservation practices, even with endangered species [10,11] It is based on the PCR amplification of discrete regions of genome with short oligonucleotide primers of arbitrary sequence [12]. The characters assessed through RAPD are useful for genetic studies because they provide various types of data-taxonomic population, inheritance pattern of various organisms including fishes [13].…”
