Glucose-regulated protein 78 is an antiviral against hepatitis A virus replication

Abstract: Infection with hepatitis A virus (HAV) is a major cause of acute hepatitis globally and it is important to identify the mechanisms of HAV replication. Glucose-regulated protein 78 (GRP78) is an endoplasmic reticulum (ER) chaperone and serves a role in unfolded protein response pathways. Previous studies have demonstrated that GRP78 functions as an endogenous antiviral factor. In the present study, two loss-of-function studies using GRP78 were completed to elucidate the role of GRP78 in HAV infection. HAV repli… Show more

“…We observed that HAV HA11-1299 genotype IIIA strain replication is enhanced by the accumulation of lipids or high-concentration glucose in the human hepatoma cell line, Huh7 [ 26 , 27 , 28 ]. Hyperglycemia or the accumulation of lipids induces an endoplasmic reticulum (ER) stress response in human hepatocytes.…”

“…GRP78 is a negative regulator of ER stress response. We also observed that the overexpression of GRP78 could inhibit HAV replication, while the knockdown or knockout of GRP78 enhanced HAV replication [ 26 , 28 ]. In sum, GRP78 is an antiviral protein against HAV replication [ 28 ].…”

“…We previously observed that HAV replication was associated with a reduction in glucose-regulated protein 78 (GRP78) expression 9 and reported that GRP78 is an antiviral against HAV infection 10 . Miso is a traditional Japanese seasoning and is used for miso soup, a Japanese culinary staple.…”
