Glycobiomarkers by glycoproteomics and glycan profiling (glycomics): emergence of functionality

Abstract: Glycans stand out from all classes of biomolecules because of their unsurpassed structural complexity. This is generated by variability in anomeric status of the glycosidic bond and its linkage points, ring size, potential for branching and introduction of diverse site-specific substitutions. What poses an enormous challenge for analytical processing is, at the same time, the basis for the fingerprint-like glycomic profiles of glycoconjugates and cells. What's more, the glycosylation machinery is sensitive to … Show more

“…In addition, the occurrence of many diseases accompanies with the glycosylation state of related proteins, thus a lot of glycoproteins have been used as disease biomarkers and therapeutic targets [2,3]. However, many glycoproteins of research and clinical importance usually exist in very low abundance while high-abundance interfering species co-exist in sample matrix.…”

“…This may be linked to viral transformation. In fact, genetic alterations are known to affect the glycome [65]. When testing a series of glycocompounds, extent of GRP-C (and C-GRP-C) binding to the chicken cells was reduced by the presence of heparin, less by other proteoglycans or sulfated polysaccharides (Fig.…”

“…Purification from extracts of tumor cells or tissue using affinity chromatography provides material for characterization and functional testing [59,60]. Given this expression of the glycobiological effectors in tumors, a new research area is opened, with the potential to identify a prognostic indicator and targets for therapy [61,62]. To test these assumptions, the developments of experimental approaches to detect lectin presence (alone and embedded in a network) and the expression of binding partners – in view of the mentioned cases for orchestration of both sides of this recognition system – are the central challenges.…”
