Growth characteristics and cytokine/chemokine induction profiles of dengue viruses in various cell lines

Abstract: Summary. -The dengue disease varies clinically from mild fever to hemorrhagic fever up to potentially fatal shock syndrome. Such differences in manifestation of dengue virus (DENV) infection have been ascribed to intrinsic differences in DENVs. Four DENV serotypes or four dengue viruses (DENV-1 to DENV-4) have been clearly distinguished; however, only limited information is available on their biological characteristics in vitro. To shed more light on this subject, replication kinetics of all four DENVs in vari… Show more

“…The permeability of the vero cell infected by dengue virus increases by the time. The leakage shown by low titer of dengue virus agrees with the result found by previous research [10 ], that reported the growth of virus number in the vero cells especially 24 hours post infection. The increase of virus number will increase the cytokines production, which in turn increases the cell permeability and causes the fluid leakage as found in the recent research.…”

“…Previous research found that in response to dengue infection, epithelial cells A549 can release 26 cytokines/chemokines including TNF-α, TNF-β, IL-1 α, IL-1 β, IL-2, IL-3, IL-4, IL-5, IL-6, IL-7, IL-8, IL-9, IL-10, IL-12(p40), IL-12(p70), IL-13, IL-15, IL-17, G-CSF, GM-CSF, IFN-α2, IFN-β, MCP-1, MIP-1α, MIP-1β and Eutaxin. Those cytokines are responsible for the increase of cell permeability [10].…”

“…Virus titer was quantified as viable virus in the supernatant using plaque assay method, as described elsewhere [10, 28]. Briefly, 24-well plates were seeded with BHK21 cells in RPMI complete medium at a density of 2 × 10 5 cells/well and incubated at 37 °C incubator, 5% CO 2 .…”

“…The role of cytokines and chemokines in the pathogenesis of severe dengue has been reported [8, 9]. Using an in vitro cell infection model, the cytokine/chemokine expression profiles of DENV-infected cells has been described [10].…”
