High performance liquid chromatography analysis of MESNA (2-mercaptoethane sulfonate) in biological samples using ?uorescence detection

Abstract: MESNA is the sodium salt of 2-mercaptoethane sulfonate, a thiol-containing drug. It is an antioxidant used particularly in renal protection. Several studies have proved that MESNA has beneficial effects in ischemic acute renal failure where it scavenges reactive oxygen species (ROS), due to the presence of the thiol group. It also reduces the size of urinary bladder cancer. MESNA was proved to be effective in preventing hemorrhagic cystitis induced by high doses of several chemotherapeutic regimens such as cyc… Show more

“…Several drugs, i.e., TP in biological samples (plasma, lung, liver, kidney and brain in mice) [63], 2-mercaptoethane sulfonate (MESNA) in biological samples (lung, liver, kidney and brain) [64], captopril in rat plasma [65], and sodium thiosalicylate in mice plasma [66], were successfully determined by the LC-FL method, as well as biological thiols [67]. The reaction scheme of ThioGlo TM 3 with captopril is shown in Fig.…”

“…ThioGlo TM 3 was used as a derivatizing agent because this reagent has several advantages such as a low fluorescence background, a high quantum yield after reacting with free thiols, a fast reaction time, and a slow hydrolysis rate of the adduct [15,16]. Furthermore, ThioGlo TM 3 has been used as a fluorescent probe, in combination with HPLC, to successfully analyze a number of free thiol substrates and drugs in biological samples such as glutathione [17], captopril [16] and MESNA [18].…”

“…Previous work carried out by the authors of this paper on the stability of Ifosfamide and Mesna in elastomeric pumps using HPLC-PDA, showed an apparent increase in Mesna concentration 5 over the duration of analysis (24,25) (26)(27)(28)31) Despite the widespread clinical use of Mesna, few studies have investigated its stability using a suitable analytical method (other than HPLC-UV) and none considered the possibility of Dimesna forming in the infusion. Therefore, the aim of this study was to evaluate the physicochemical stability of Mesna and quantify any Dimesna forming at 37ºC for average adult doses; using a validated Liquid Chromatography-Mass Spectrometry (LC-MS/MS) method and Nuclear Magnetic Resonance Spectroscopy (NMR).…”
