Histidine inhibits oxidative stress‐ and TNF‐α‐induced interleukin‐8 secretion in intestinal epithelial cells

Abstract: We investigated the effect of several amino acids on the secretion of such inflammatory cytokines as interleukin-8 (IL-8) induced by hydrogen peroxide or tumor necrosis factor-alpha (TNF-a) in intestinal epithelial-like Caco-2 and HT-29 cells. We found that histidine, one of the conditionally essential amino acids, significantly inhibited both hydrogen peroxide-and TNFa-induced IL-8 secretion and mRNA expression in Caco-2 cells and HT-29 cells. These inhibitions were dose dependent and the inhibition rate of h… Show more

“…2C shows that IFF did not affect hydrogen peroxide-induced IL-8 induction, although hydrogen peroxide also induces IL-8 secretion via NFB activation (22). It has been reported that such food components as chlorogenic acid, caffeic acid, and histidine suppressed not only TNF-␣-induced IL-8 secretion but also hydrogen peroxide-induced IL-8 secretion (13,15). However, there have been no reports about food components which can suppress TNF-␣-induced IL-8 activation, but not hydrogen peroxide-induced activation.…”

“…It is therefore important to regulate and maintain the secretion of IL-8 by intestinal epithelial cells to prevent the intestinal inflammatory loop from developing. It has recently been discovered by using intestinal epithelial cell lines that such food components as flavonoids and amino acids suppressed stimulant-induced IL-8 secretion (12)(13)(14)(15). We focused the present study on the effect of food-derived components on stimulantinduced IL-8 production by intestinal epithelial cells.…”

“…Soy isoflavone reduces the secretion of TNF-␣-induced IL-8, but not of hydrogen peroxide-induced IL-8 (34). Histamine inhibits TNF-␣-and oxidative stress-induced IL-8 secretion in Caco-2 cells (35). Orally administered caffeine decreases pro-inflammatory cytokines (TNF-␣ and IL-17F) and increases anti-inflammatory cytokines IL-10 in mouse colons (36).…”

“…Histidine, a conditionally essential amino acid, significantly inhibited both H 2 O 2 -and TNF--induced IL-8 secretion and mRNA expression in Caco-2 cells and HT-29 cells. 92) TNFincreased the transcriptional activity of the IL-8 promoter, which was significantly inhibited by treating Caco-2 cells with histidine. Histidine also abolished the NF-B-dependent activation of the IL-8 promoter induced by TNF- (Fig.…”
