HLA‐B*4703: sequence confirmation, serology and distribution

Abstract: This study has confirmed the nucleotide sequence of exons 2 and 3 of the B*4703 allele, discovered by an unusual HLA-B47 and Bw6 serological pattern, in two subjects of Black/Japanese and Caribbean Black descent. Titration studies on 25 HLA-B47 cross-reactive sera, stimulated by B13, B27, B44 and B60, and nine Bw6 antisera/monoclonal antibodies, showed that the B*4703 product can be distinguished from the established HLA-B47 specificity. The phenotypes of these donors and an International Cell Exchange donor s… Show more

“…These findings strengthen our view that, even in the era of DNA‐based typing, the careful evaluation of serological reaction patterns can be fruitful in the identification of new and rare alleles ( 19–21). They also suggest that the HLA‐A2403 specificity should now be appended to other HLA‐A*24 alleles.…”

“…Nucleotide sequencing of B*4408 was performed using PCR‐generated templates from allele‐specific primer mixtures for exon 2 [containing primers 5Bin1‐TA and R219 (5′‐CCAGGTATCTGCGGAGCG‐3′)] and exon 3 [containing primers L243 (5′‐CGCGAGTCCGAGGATGGC‐3′) and 3Bin3‐37 (Cereb & Yang, 1997)] as previously described (Darke et al ., 1999; Guttridge et al ., 1999).…”

“…During routine ‘low resolution’ (minimum of ‘2‐digit” allele level) five‐locus HLA typing of patients, and blood donors enrolling on the Welsh Bone Marrow Donor Registry (WBMDR) (4) we identified two subjects who gave unexpected amplification patterns by polymerase chain reaction using sequence specific primers (PCR‐SSP). Nucleotide sequencing of exons 2 and 3 (5, 6) of the aberrant HLA‐A and B alleles identified two new class I alleles – A*0108 and B*0431 1 .…”
