Treatment of diabetic patients with antioxidant, such as extra virgin olive oil (EVOO), may be beneficial in numerous debilitating complexities. This study was aimed at assessing the protective role of virgin olive oil in reducing hyperglycemia in streptozotocin- (STZ-) induced diabetic rats. Thirty-six healthy male Sprague-Dawley rats were divided into six groups (6 rats per group) including nondiabetic control (NC), diabetic control (DC), and animals treated with metformin, olive oil, and a combination of olive oil and metformin, respectively. The protective effect of olive oil was evaluated by determining the biochemical parameters (lipid profile, liver, and kidney) and by studying the histopathological alterations in pancreas, liver, and kidney tissues. The results showed a significant increase in alanine aminotransferase (ALT) and alkaline phosphatase (ALP) levels in diabetic rats. ALP levels remained significantly elevated in the diabetic rats that were treated with metformin and/or olive oil, and the highest level was noted in the group treated with olive oil (568.33 U/L). Contrarily, pretreatment with olive oil significantly decreased ALT (67.64 U/L) and ALP (226.17 U/L) levels. Histopathological data revealed that all the disorganized islets of Langerhans along with the clusters of inflammatory cells were absent in the group pretreated with the combination of virgin olive oil and metformin, which shows that prophylactic administration of this combination reduces the diabetic complications in a much better way. Therefore, pretreatment with olive oil with or without metformin is an encouraging approach for diabetes therapy with immense potential.

Objective: The objective of the study was to evaluate the genotoxic and mutagenic potentials of Kalanchoe pinnata alcoholic leaves extract.Methods: Single cell gel electrophoresis (COMET assay) was adopted to investigate genotoxicity potential of standardized ethanolic leaves extract at the concentration of 10, 50, and 100 µg/ml, respectively, using human lymphocytes.Results: K. pinnata leaves extract did not completely damage DNA in human lymphocytes but low, medium, and high-level damage was found in with positive control. The DNA damage level decreases in low, medium, and high (12±1, 6±1, and 1±1) without positive control at the concentration of 100 µg/ml. Conclusion:It was concluded that the parent material did not induce mutagenic and genotoxic potentials at the concentrations 10, 50, and 100 µg/ml but its metabolites are inducing genotoxicity in various models.

Background:Diabetes mellitus (DM) continues to pose a major global health threat with serious economic burden in sub-Saharan Africa, especially Nigeria. This condition is often worsened with accompanying comorbidities, further putting a strain on patient wellbeing. Medicinal plants are not only effective in DM, but some of them are also have beneficial effects on accompanying disease states. Such plants may be appropriate alternatives or adjuncts to available antidiabetic medications. This study aimed at accessing and documenting the medicinal plants used in four local government of southwestern Nigeria for the management of diabetes and its comorbidities.Methods:Semi-structured questionnaires were used to obtain information from traditional medical practitioners, village elders and herb sellers. The data obtained were analyzed and discussed in relation to previously published literature. Fifty-three respondents mostly males (77.4%) provided information on medicinal plants useful for the management of diabetes and its comorbidities.Results:A total of 77 medicinal plants belonging to 76 genera in 44 plant families were mentioned in the survey. In particular, 53 medicinal plants are mentioned for managing diabetes, 35 for hypertension, 28 for stroke, 17 for heart diseases and 25 for obesity. The most frequently cited plant family was Leguminosae (12%), followed by Euphorbiaceae (8%). The frequently used plant species includesVernonia amygdalina, Citrus aurantifolia, Viscum album, Carica papaya, Allium sativum. It is interesting to note that only Citrus aurantifoliawas mentioned severally for the management of the five ailments. The herbal recipes, methods of herbal preparation and administration were also recorded.Conclusions:The survey shows that a significant number ofmedicinal plants are used for the management of diabetes and its comorbidities in the study area. These plants, apart from their hypoglycemic activity, may be useful against the most common complications of DM and may provide strategies for the development of less expensive therapies especially in low income communities.
