In vitro cytotoxicity of maxillofacial silicone elastomers: Effect of accelerated aging

Abstract: The purpose of this in vitro study was to evaluate the cytotoxicity of three maxillofacial silicone elastomers at 24, 48, and 72 h on L-929 cells and to determine the effect of accelerated aging on the cytotoxicity of these silicone elastomers. Disc-shaped test samples of maxillofacial silicone elastomers (Cosmesil, Episil, Multisil) were fabricated according to manufacturers' instructions under aseptic conditions. Samples were then divided into three groups: (1) not aged; (2) aged for 150 h with an accelerate… Show more

“…Other researchers have studied the biocompatibility of room-temperature silicone elastomers using the MTT assay, 42,43 and concluded that cell survival in the presence of silicone elastomers was affected by incubation periods. In our current study, we found that cell survival increased proportionally with the length of the incubation period.…”

“…The MTT test is considered to be a sensitive quantitative method of evaluating cell viability and proliferation in vitro [14,15]. Yellow tetrazolium salt (MTT) is reduced to purple formazan in the living cells or cells in early apoptosis cycle with active mitochondrial dehydrogenases [16,17].…”

“…Since silicone elastomer was first successfully used as a prosthetic material in 1960, it has become the first choice maxillofacial prosthetic material for soft tissue repair . Silicone elastomers have been generally used to restore facial prostheses because of their chemical and mechanical properties, strength, ease of manipulation, pigmentability, and durability . With regard to ideal qualities (chemical, physical, and mechanical) of a prosthetic material, it should be compatible with human tissues and cause no irritation .…”
