In Vivo Coinstantaneous Identification of Hepatocellular Carcinoma Circulating Tumor Cells by Dual-Targeting Magnetic-Fluorescent Nanobeads

Abstract: Circulating tumor cells (CTCs) have been considered as a potential biomarker for evaluation of cancer metastasis and prognosis, especially in hepatocellular carcinoma (HCC). However, the isolation and detection of rare CTCs in HCC patients face enormous challenges due to omittance and nonspecific binding. We previously designed a small molecular NIR fluoresent agent, named MLP, which had high affinity with a tumor cell-overexpressed enzyme, aminopeptidase N (APN). Based on that, in this work we introduced a no… Show more

“…For example, EpCAM-based immunomagnetic enrichment followed by multiparameter flow cytometric and immunocytochemical analysis has been developed for precise quantification of CTC number in cancer patients (56). Various anti-EpCAM and anti-ASGPR monoclonal antibody (mAb)-based platforms for phenotypic profiling and isolation of HCC-derived CTCs in HCC patients have been proposed (57,58).…”

“…As the most frequent one among primary hepatic cancers (75–85%), hepatocellular carcinoma (HCC) exhibits a high recurrence rate of 60–70% in 5 years, and the highly sensitive diagnosis and efficient therapy on HCC in the early stage are crucial to the suvival. , Among all clinical therapies for patients suffering from HCC, hepatectomy and transcatheter arterial chemoembolization (TACE) were dominant. Single clinical treatment was recognized to be almost impossible to cure HCC, so multidisciplinary treatment was highly recommended to treat HCC in the Clinical Practice Guidelines . In particular, new materials and combined therapy were highly demanded to improve the therapeutic efficacy for HCC.…”

“…[4][5][6][7] Among these developments, IMNs have been found to be the most extensively used approach due to their fast isolation and simple operation. 8,9 However, there are still some challenges related to the clinical application of IMNs, such as conforming to capture efficiency and purity requirements for separating CTCs from a complex blood environment. 10,11 When the nanoparticles enter body fluids, proteins spontaneously adsorb onto the particles to form a protein corona.…”
