Metascardovia criceti Gen. Nov., Sp. Nov., from Hamster Dental Plaque

Abstract: A novel microorganism, Metascardovia criceti gen. nov., sp. nov., was isolated from dental plaque of golden hamsters fed with a high‐carbohydrate diet. The three isolated strains, OMB104, OMB105, and OMB107, were Gram‐positive, facultative anaerobic rods that lacked catalase activity. Analyses of the partial 16S rRNA and heat‐shock protein 60 (HSP60) gene sequences of these isolates indicated that they belonged to the family Bifidobacteriaceae. However, in contrast to Bifidobacterium, one of the genera under t… Show more

“…In agreement with previous culture-based studies, Bifidobacterium species are underabundant in T-bet -/- × Rag2 -/- mice [68], and LEfSe highlights several additional genus-level clades, including the specifically depleted Roseburia and Papillibacter within the otherwise overabundant Firmicutes.…”

“…At the family level, the Rag2 -/- enrichment of Bifidobacteriaceae, Porphyromonadaceae, Staphylococcaceae and the T-bet -/- × Rag2 -/- enrichment of Lachnospiraceae confirm our reports in [68] using culture-based and quantitative real time PCR techniques. LEfSe's LDA score more informatively reorders these taxa relative to the P -values found for these families in our previous work, highlighting the Bifidobacteria and, interestingly, several clades within the Clostridia.…”

“…Phylogenetic markers useful in the taxonomy of the family Bifidobacteriaceae, partial sequences of the hsp60, fusA, xfp, tuf, rpoC, rplB, gyrB and dnaK genes, were considered. A fragment of the hsp60 gene was amplified from the studied strain using primers MO-157 and MO-158 under PCR conditions described previously (Okamoto et al, 2007). PCR conditions and primers for amplification of fusA, rplB and gyrB gene sequences were taken from Delétoile et al (2010).…”

“…The hsp60 gene has been proposed as an alternative phylogenetic marker for the family Bifidobacteriaceae (Jian et al, 2001;Huys et al, 2007). Hence, partial sequences (~590 bp) of the hsp60 gene were determined as described by Okamoto et al (2007). Fragments were then sequenced on a 3100-Avant Genetic Analyzer with a BigDye Terminator version 3.1 cycle sequencing kit (both from Applied Biosystems).…”
