Sinorhizobium melilotistrain 1021bioSandbdhAgene transcriptions are both affected by biotin available in defined medium

Abstract: Sinorhizobium meliloti 1021 responds to external biotin signals from alfalfa plants through the bioS regulatory locus. Immunogold labeling and electron microscopy revealed that the BioS protein is located within the S. meliloti cytoplasm. Under biotin-limiting conditions the S. meliloti cell lumen was filled with polyhydroxybutyrate (PHB) granules suggesting that either PHB synthesis or degradation are influenced by biotin. To test this hypothesis a 3-hydroxybutyrate-dehydrogenase-lacZ (bdhA-lacZ) fusion was m… Show more

“…It has been reported that PHB was not accumulated in the free-living culture of R. etli when biotin was added [90]. The transcription of bdhA in free-living S. meliloti was higher in the presence of added biotin [91]. These earlier reports suggested biotin-induced PHB degradation in the free-living condition.…”

“…Also in this region is SMa0669 , which is predicted to encode an HlyD family protein that contains a biotin binding domain. Biotin has previously been associated with PHB accumulation ( 64 ).…”

“…Beside its role as a cofactor in carboxylation reactions, biotin plays a pivotal role during root colonization in sinorhizobia (71). It is of importance for the synthesis and degradation of polyhydroxybutyrate (72) and various anaplerotic reactions in rhizobia and bradyrhizobia (73,74). A minimum set of six genes is generally required for biotin biosynthesis in Gram-negative bacteria, and they are usually organized within a single transcriptional unit (75).…”
