Identification of a Flavonoid Glucosyltransferase Involved in 7-OH Site Glycosylation in Tea plants (Camellia sinensis)

Abstract: Flavonol glycosides, which are often converted from aglycones in a process catalyzed by UDP-glycosyltransferases (UGTs), play an important role for the health of plants and animals. In the present study, a gene encoding a flavonoid 7-O-glycosyltransferase (CsUGT75L12) was identified in tea plants. Recombinant CsUGT75L12 protein displayed glycosyltransferase activity on the 7-OH position of multiple phenolic compounds. In relative comparison to wild-type seeds, the levels of flavonol-glucosides increased in Ara… Show more

“…In addition to the characteristics described above, the pH optima of the recombinant enzyme seem unlikely to be of physiological importance in vivo . But similar results have been reported in rCsUGT75L12 and CsUGT73A20 from tea plants, and UGT78K1 from black soybean showed high pH optima in in vitro biochemical assays, but was demonstrated to play significant roles in the biosynthesis of flavonoids in vivo ( Kovinich et al, 2010 ; Dai et al, 2017 ; Zhao et al, 2017 ). These UGTs show significant enzyme optima; as such, they are good candidates to engineer flavonoid diversity.…”

“…The 3- O -glycosylation of flavanols (kaempferol, quercetin) is catalyzed by the UGT78D family, with UGT78D1 using UDP-rhamnose and UGT78D2 using UDP-glucose [ 48 ]. The recently identified gene-encoding flavonoid 7- O -glycosyltransferase ( CsUGT75L12 ) in tea plant displays glycosyltransferase activity on the 7-OH position of multiple phenolic compounds [ 49 ]. In the present work, we observed low levels of m-trigallic acid and 2,4-dihydroxybenzoic acid, a finding in agreement with previous ones of decreased benzoids in P-starved Arabidopsis [ 10 ].…”

“…The binary vector was introduced into A. tumefaciens EHA105 cells through electroporation. A positive colony was injected into N. benthamiana leaves, as reported previously (Dai et al , ). The infected leaves were cultivated in the glasshouse for 48 h and subsequently examined under an Olympus FV1000 confocal microscope.…”
