Identification of alien chromatin specifying resistance to wheat streak mosaic and greenbug in wheat germ plasm by C-banding and in situ hybridization

Abstract: The chromosome constitutions of eight wheat streak mosaic virus (WSMV)-resistant lines, three of which are also greenbug resistant, derived from wheat/ Agropyron intermedium/Aegilops speltoides crosses were analyzed by C-banding and in situ hybridization. All lines could be traced back to CI15092 in which chromosome 4A is substituted for by an Ag. intermedium chromosome designated 4Ai-2, and the derived lines carry either 4Ai-2 or a part of it. Two (CI17881, CI17886) were 4Ai-2 addition lines. CI17882 and CI17… Show more

“…However, modern breeding was challenged by the narrow genetic variation [1,2], which affected further improvements in wheat yield and quality. Wild relatives of wheat possessed many desirable and valuable traits that could be used as gene resources for wheat improvement [3][4][5]. For example, Agropyron cristatum chromosome 6P with the genes controlling large numbers of florets and kernels per spike and multiple fertile tiller numbers per plant [6,7]; Dasypyrum villosum chromosome 1V including the seed storage…”

“…Fluorescence in situ hybridization (FISH) is well established as a highly efficient method for cytological characterization. It has been used for physical mapping of specific sequences in wheat and its relatives (Mukai et al, 1990 and1992;Leitch et al, 1991; to locate the breakpoints of chromosome translocations (Friebe et al, 1991;Mukai et al, 1993;, Molnar-Lang et al, 2000 and in karyotyping (Cabrera et al, 1995;Tsujimoto et al, 1997).…”

“…Genetic resistance to WSMV offers the most environmentally and economically desirable strategy for disease control and has been pursued since resistance to WSMV was discovered (Friebe et al, 1991;Graybosch et al, 2009;Mutti et al, 2011). Today, there is huge interest in both using genomic prediction to access genotype × environment interaction using marker and pedigree information in breeding programmes (Crossa et al, 2014), and in enhancement of the genetic variability available in wheat through wide hybridisation (see Heslop-Harrison and Schwarzacher, 2012).…”
