Identification of avocado (Persea americana) pulp proteins by nano‐LCMS/MS via combinatorial peptide ligand libraries

Abstract: Avocado (Persea americana) proteins have been scarcely studied despite their importance, especially in food related allergies. The proteome of avocado pulp was explored in depth by extracting proteins with capture by combinatorial peptide ligand libraries at pH 7.4 and under conditions mimicking reverse-phase capture at pH 2.2. The total number of unique gene products identified amounts to 1012 proteins, of which 174 are in common with the control, untreated sample, 190 are present only in the control and 648 … Show more

“…The protocols are summarized in Fig. , as based on previously published works . In a first extraction method, performed by using physiological conditions, 5 g of Goji berries were ground and homogenized with 5 mL of a buffer containing 0.125 M Tris‐HCl (pH 7.4) and protease inhibitor cocktail.…”

“…In this field, the use of combinatorial peptide ligand libraries (CPLLs) has demonstrated to be a very powerful tool. The efficiency of CPLLs in the capture of low to very‐low abundance proteins has been widely demonstrated in the treatment of a number of samples , but it was just in the last few years when this technology has been applied to the study of vegetable samples such as maize kernels , spinach leaves , olive , avocado , and banana . Generally, the increment of identified species has been between 200 to 500% as compared with the nontreated control samples.…”

“…Also, changes of sugars, total protein and oil in 'Hass' avocado have been determined using GC-FID and LC [54]. On the other hand, important proteins from the pulp have been identiied through the use of nano-LC-MS/MS [55]. The use of coupled analytical systems has helped to get beter resolution, higher sensitivity, higher speed of analysis and wider diversity of applications.…”

“…This is a tropical fruit largely consumed in various parts of the world as vegetable alone or with salads. In the context of low-abundance gene product discovery, a paper has been recently published (27). Avocado is also used as a source of products for cosmetics and healthcare (45).…”
