Identification of serum biomarkers for lung cancer using magnetic bead-based SELDI-TOF-MS

Abstract: Aim: To identify novel serum biomarkers for lung cancer diagnosis using magnetic bead-based surface-enhanced laser desorption/ionization time-of-flight mass spectrum (SELDI-TOF-MS). Methods: The protein fractions of 121 serum specimens from 30 lung cancer patients, 30 pulmonary tuberculosis patients and 33 healthy controls were enriched using WCX magnetic beads and subjected to SELDI-TOF-MS. The spectra were analyzed using Biomarker Wizard version 3.1.0 and Biomarker Patterns Software version 5.0. A diagnostic… Show more

“…Recent biomarker discovery programs aimed at identifying novel TB biomarkers with demonstrable utility have been successful [17]–[19]. Among the analytical tools for TB biomarker discovery, transcriptional microarray [17], [20], [21], 2D gel analysis [22], and mass spectroscopy [18], [23] have been used but none, to our knowledge, has undertaken such a large-scale analysis for the targeted detection of protein levels in the serum of patients with TB disease.…”

“…Although both are weakly cationic, as a new protein separation technology, the WCX magnetic beads provide a great flexibility for fractionation of complex biological samples. It has been successfully used in the separation and purification of various proteins in the body fluids such as serum, plasma and amniotic fluid [20][21][22]. During the pilot study, we compared the SELDI spectra of the serum in TB patients by WCX magnetic beads and CM10 protein chips.…”

“…Nevertheless, a number of peaks that we identified, for example, the peaks M/Z 1205and 4279, were not recognized previously. 32,33 This is conceivably due to the different experimental procedures used in the studies. Moreover, the varying approaches in data analysis may also contribute to the difference.…”
