IGF-II promoter methylation and ovarian cancer prognosis

Abstract: This study shows that methylation varies among three IGF-II promoters in ovarian cancer and that this variation seems to have biologic implications as it relates to clinical features and prognosis of the disease.

“…Methylation-specific PCR and bisulphite sequencing analysis of IGF2 promoter regions Genomic DNA from tumour and normal liver samples was treated with sodium bisulphite (Herman et al, 1996), and the methylation status of the P2 -P4 promoter regions of IGF2 was analysed by methylation-specific PCR (MSP), as described earlier (Beeghly et al, 2007). Polymerase chain reaction products were run on 2% agarose gels and visualised after staining with ethidium bromide.…”

“…Exons 8 to 10 encode the mature protein, thus each transcript generates the same IGF-II peptide [162]. However the four promoters (under control of different transcription factors), and six transcripts (with varying 5' untranslated regions, translatability and stability), regulate the spatial, temporal and level of protein expression [177]. IGF-II is induced by hypoxia in HepG2 human hepatoma cells via its P3 promoter, upregulation of the transcription factor Egr-1 and enhanced binding of Egr-1 to the IGF-II promoter [176].…”

“…HTX embryos expressed less (<1 fold) Igf2 than the controls; apparently this difference does not affect embryo size. This suggests that other Igf2 regulating elements such as the Igf2 promoter [Beeghly et al 2007] may also be required to differentially regulate the HVPX and HTX groups.…”
