Impact of different culture media on hairy roots growth of Valeriana officinalis L.

Abstract: Transformed hairy root cultures of Valeriana officinalis were established by infection with Agrobacterium rhizogenes strain ATCC 15834. To determine the effect of different media on the growth of V. officinalis hairy roots, MS, B5 media (1.0X and 0.5X strength), N6 medium and a modified MS medium without phytohormones were used. In addition, different NH 4 + to NO 3 -ratios in MS medium were studied. The effects of these treatments were evaluated after 21 days of culture in relation to hairy root growth. B 5 a… Show more

“…These results were in line with previously published studies, e.g. B5 and ½ B5 media were the best basal media for hairy root growth of V. officinalis (Pakdin et al, 2014) and the NB medium composition supported best growth of hairy roots in Glycyrrhiza glabra followed by MS, B5 and WP media (Mehrotra et al, 2008). In G. glabra 20 times increase in root biomass on fresh mass basis was recorded after 45 days of culture in NB medium.…”

“…Different concentrations of salts and elicitors in culture media have a major role in hairy root growth and induction of secondary metabolites production (Wang and Wu, 2013). It is worth to mention that hairy roots growth drastically increase by change in some major mineral components (Pakdin et al, 2014).…”

“…As for the FW and DW, anthocyanin concentration was lower for HRs grown on full MS. On the other hand, anthocyanin concentration was higher for both HR lines grown on 1/2 MS+S, and for NB-R grown on 1/2 MS (Figure 2c). In this sense, an excess of nutrients could redound negatively on HR growth under our experimental conditions; this has been reported for HRs and the tissue culture of several species, where higher nutrient content in the medium either decreased or did not affected biomass accumulation [25,39,40]. Anthocyanin yield, expressed as µg anthocyanin per experimental unit (plate), can be an adequate reference to high-productive experimental conditions, as it takes into account both biomass and anthocyanin concentration.…”
