Introduction: The current study was designed to synthesize derivatives of succinimide and compare their biological potency in anticholinesterase, alpha-glucosidase inhibition, and antioxidant assays. Methods: In this research, two succinimide derivatives including (S)-1-(2,5-dioxo-1-phenylpyrrolidin-3-yl) cyclohexanecarbaldehyde (Compound 1) and (R)-2-((S)-2,5-dioxo-1-phenylpyrrolidin-3-yl)-2-phenylpropanal (Compound 2) were synthesized using Michael addition. Both the compounds, ie, 1 and 2 were evaluated for in-vitro acetylcholinesterase (AChE), butyrylctcholinesterase (BChE), antioxidant, and α-glucosidase inhibitory potentials. Furthermore, molecular docking was performed using Molecular Operating Environment (MOE) to explore the binding mode of both the compounds against different enzymes. Lineweaver-Burk plots of enzyme inhibitions representing the reciprocal of initial enzyme velocity versus the reciprocal of substrate concentration in the presence of synthesized compounds and standard drugs were constructed using Michaelis-Menten kinetics. Results: In AChE inhibitory assay, compounds 1 and 2 exhibited IC 50 of 343.45 and 422.98 µM, respectively, against AChE enzyme. Similarly, both the compounds showed IC 50 of 276.86 and 357.91 µM, respectively, against BChE enzyme. Compounds 1 and 2 displayed IC 50 of 157.71 and 471.79 µM against α-glucosidase enzyme, respectively. In a similar pattern, compound 1 exhibited to be more potent as compared to compound 2 in all the three antioxidant assays. Compound 1 exhibited IC 50 values of 297.98, 332.94, and 825.92 µM against DPPH, ABTS, and H 2 O 2 free radicals, respectively. Molecular docking showed a triple fold in the AChE and BChE activity for compound 1 compared with compound 2. The compound 1 revealed good interaction against both the AChE and BChE enzymes which revealed the high potency of this compound compared to compound 2. Conclusion: Both succinimide derivatives exhibited considerable inhibitory activities against cholinesterases and α-glucosidase enzymes. Of these two, compound 1 revealed to be more potent against all the in-vitro targets which was supported by molecular docking with the lowest binding energies. Moreover, compound 1 also proved to have antiradical properties.

Ankylosing spondylitis (AS) is a chronic, progressive, and inflammatory disease that mainly affects the central axis joint. Although this disease has already been well documented and studied, its pathogenesis is still not well understood. This study aimed to screen and identify key candidate genes involved in the progression of AS. For this purpose, expression profiles of GSE39340 and GSE41038 were downloaded from the Gene Expression Omnibus and displayed in the form of volcano plots and heatmaps. Differentially expressed genes (DEGs) were identified by the Limma package in R and functional enrichment analyses were performed. Moreover, STRING and Cytoscape were utilized to construct protein-protein interaction (PPI) networks and screen significant modules. Immunohistochemistry (IHC) in tissue chips of AS and normal human synovial tissues was performed to confirm the major proteins associated with its development. Western blotting (WB) and alizarin red staining were applied to validate the expression level of platelet-derived growth factor receptor beta (PDGFRB) and function during osteogenesis differentiation of fibroblasts in AS. A total of 256 DEGs were screened, including 191 up-regulated genes and 65 down-regulated genes. The enriched functions of these identified genes mainly included adherens junction, focal adhesion, and cell-substrate adherens junction. The pathways most highly associated with the progression of AS were TGF-β signaling pathway, the Hippo signaling pathway, and the AGE-RAGE signaling pathway. In addition, IHC showed that mitogen-activated protein kinase 1 (MAPK1), C-X-C motif chemokine receptor 4 (CXCR4), and PDGFRB were highly expressed in AS. PDGFRB was found upregulated during osteogenesis of fibroblasts and stimulates osteogenesis in AS. These findings may improve our understanding of the molecular mechanisms controlling AS. Pharmacological targeting of PDGFRB may initiate a possible suppression of bone formation in AS.

Medicinal plants are potential natural sources of biologically active substances that have geroprotective properties, slowing down the aging process. Vegetable raw materials are used as antioxidants in the food industry. Pulmonaria officinalis L. has a high content of biologically active substances. The purpose of this work is to determine the operating parameters for extracting the maximum amount of biologically active substances from Pulmonaria officinalis L. by extracting ethanol and finding sources of natural antioxidants.The antioxidant activity of several ethanol extracts of Pulmonaria officinalis L. – 30-70% with a step of 10 ethanol obtained from the dried root culture of the medicinal plant of biologically active substances was determined. The total content of polyphenols, flavonoids and proanthocyanidins was estimated. According to the data obtained, the relationship between the values determined for the biologically active substances presented and the values of antioxidant activity was revealed. Extracts of Pulmonaria officinalis L. showed a high yield of polyphenolic compounds-889.39±4.29 mcg of Gallic acid/ml, flavonoids-728.90±6.98 mcg of rutin/ml, proanthocyanidins-211.65±5.31 mcg of catechin/ml at an ethyl alcohol concentration of 60% and an extraction temperature of 50°C in 4 hours. The antioxidant activity using 2 methods: A spectrophotometric method DPPH (2,2-diphenyl-1-picrylhydrazyl) and the FRAP method (iron reducing/antioxidant power) was investigated. During DPPH spectrophotometric analysis, the antioxidant activity is equal to 86.96% for polyphenols, 75.47% for flavonoids, and 51.25% for proanthocyanidins. FRAP analysis showed that extracts with operating parameters had a pronounced antioxidant activity (tэ= 4 ч; Tэ= 50°C; Сэ= 60%)и (tэ= 5 ч; Tэ= 60°C; Сэ, = 70%).
