A spectrophotometric investigation has been made of the effect of particulate solids on the activity of selected antimicrobial preservatives. By subjecting these systems to high temperatures and pressures it was possible to determine whether any decrease in activity was due to absorption or chemical degradation of the respective preservative present. Qualitative and quantitative procedures for the analysis of creams of the oil-in-water type are reviewed and discussed, with special attention to the clean-up methods. It is concluded that systematic investigations have hardly been reported, except for creams containing corticosteroids.It5-I98O Pharmaceutisch Weekblad I429 ABSTRACT A simple and rapid method for the spectrofluorimetric determination of adrenaline and some related compounds in pharmaceutical preparations was developed. The method is based on the measurement of the fluorescence of the compounds at their respective emission maxima after excitation of solutions in o. I M sodium dihydrogen phosphate. The influence of several additives (drugs, preservatives, buffers, etc.) was studied. With the method amounts of 1-4 /zg/ml of the various drugs could be determined. The method was tested with a variety of pharmaceutical formulations.

The uv absorbing properties of the components of the cream bases as described in the Formulary of the Dutch Pharmacists were investigated. Direct ov spectrophotometric determinations without any clean-up steps appeared to be possible for a number of drugs (e.g. tripelennamine HCI, tretinoin, salicylic acid, methyl salicylate, resorcinol, clioquinol), with the help of a solvent mixture in which the cream samples dissolved completely to yield clear solutions. Correcting for the contribution to the uv absorbance by the preservative is sometimes necessary and can be achieved by measuring the absorbance at two wavelengths. The determination of chlorhexidine, as an example of a basic drug with uv absorbing properties which prevent direct measurements of the solution of the cream samples, could be achieved after removal of the interfering compounds by a simple liquid-liquid extraction.
