Increased numbers of total nucleated and CD34+ cells in blood group O cord blood: an analysis of neonatal innate factors in the Korean population

Abstract: CB with blood group O has unique hematologic variables in this large-scale analysis of Korean neonates, although the impact on the storage policies of CB banks or the clinical outcome of transplantation remains to be determined.

“…ABO blood type is associated with several circulating markers, including von Willebrand factor (vWF), factor (F) VIII [1 -6] , E-selectin [7] , P-selectin, intercellular adhesion molecule-1 [8] and CD34-positive cells [9] . FVIII and vWF have important clinical implications in that their plasma levels are closely related with bleeding or thrombotic disorders.…”

“…We previously reported the relationship between ABO blood group and hematopoietic parameters (TNC count, CD34+ cell count, and CD34+ cells/TNCs) in CB units [5]. CB obtained from neonates with blood type O has higher TNC and CD34+ cell counts and ratio of CD34+ cells to TNCs than neonates with other blood types; therefore, CB units with blood type O may have greater hematopoietic potency.…”

“…MPV was calculated by dividing the plateletcrit by the number of platelets. After volume reduction for cryopreservation, the blood was further tested to determine TNC and CD34+ cell counts and ABO blood group, according to the standard operating procedures of Allcord as described elsewhere [5]. …”

“…CBUs were collected after obtaining informed consent from the mother and were processed for cryopreservation according to the standard operating procedures of Seoul Metropolitan Government Public Cord Blood Bank (Allcord) [34]. The collection bag weight was measured, and the blood volume was calculated with the density factor of 1.05 g/mL.…”

“…After thawing, the cord blood was poured into a test tube by incising the ends of the capillaries, and the cord blood variables (TNC and CD34 + cell count, cell viability test, and CFU assay) were assessed using previously described methods [23]. In this study, the first capillary was thawed and analyzed after one year of storage.…”
