Individual variation in contractile cost and recovery in a human skeletal muscle.

Abstract: This study determined the variation among individuals in ATP use during contraction and ATP synthesis after stimulation in a human limb muscle. Muscle energetics were evaluated using a metabolic stress test that separates ATP utilization from synthesis by 31P NMR spectroscopy. Epicutaneous supramaximal twitch stimulation (1 Hz) of the median and ulnar nerves was applied in combination with ischemia of the ringer and wrist flexors in eight normal subjects. The linear creatine phosphate (PCr) breakdown during is… Show more

“…PCr recovery is a useful measure of skeletal muscle oxidative capacity and is dependent on mitochondrial respiratory function (3,11). A further advantage of the measure is that PCr recovery does not require a correction for active muscle mass (10,16) and is independent of the work level (17), provided that muscle intracellular pH does not fall severely (2).…”

“…The calf-slope was determined for calf plantar flexion and the arm-slope for forearm flexion. Because standardized PCr decreases linearly in response to a progressively increasing work rate, 15,27,28 the PCr slope is a simple indicator of the rate of PCr breakdown against the work rate, which may mainly reflect the oxidative capacity of skeletal muscle. We used the muscle pH at the submaximal work rate to evaluate muscle acidification.…”

“…No correction for partial saturation was made, assuming that T 1 relaxation of PCr and P i remained constant for each resonance throughout the trials. Data were then expressed as the ratios of PCr/(PCr ϩ P i ) to correct signal loss or movement artifacts during the procedure (18). Intracellular pH was calculated from the chemical shift of P i based on the equation (3,19): pH ϭ 6.75 ϩ Log [(␦ -3.27)/(5.69 -␦)], where ␦ is the chemical shift of the P i peak (in ppm) with respect to PCr.…”
