“…Higher concentrations of 6-BA (10 mg·L −1 ) repressed betacyanin content and callus growth of S. salsa. The results indicate that the increase in betacyanin content with a combination of 6-BA and IAA is related to vigorous growth, which is consistent with such reports in seedlings (Piattelli and Imperato 1971) and calluses of Amaranthus tricolor (Bianco-Colomas and Hugues 1990), in cell suspension cultures of P. Americana (Hirano and Komamine 1994), and in Portulaca (Noda and Adachi 2000). Bhuiyan and Adachi (2003) also showed that cell growth and the accumulation of betacyanin were positively correlated.…”

“…For example, cytokinins increase betacyanin accumulation in Chenopodium album (Rudat and Göring 1995), Saussurea medusa , and Alternanthera brasiliana (Silva et al 2005), but addition of cytokinin decreases betacyanin accumulation in cell suspension cultures of Portulaca (Noda and Adachi 2000) and Mammillaria candida (Santos-Diaz et al 2005). The exact mechanism of regulation by growth regulators in betacyanin metabolism remains unclear.…”

“…Surprisingly, our primary results showed that light enhanced the betacyanin accumulation in S. salsa calli. Previous studies have shown that cell cultures can be a useful tool for analyzing the regulatory mechanism of betalain biosynthesis and for the characterization of the enzymes involved (Noda and Adachi 2000). In the present study, we introduced the C 3 halophyte S. salsa calli as a model system for studying the effect of light on betacyanin content and whether there is a correlation between DODA expression or activity and expression of tyrosinase and betacyanin content in the calli of S. salsa under light.…”

“…Glucose concentration was measured using a glucose analyzer (model 2700, YSI Inc., Yellow Springs, OH, USA). Alanine concentration was analyzed by high-performance liquid chromatography (HPLC) as described previously (Meyer et al 1984;Noda and Adachi 2000) with modifications. Alanine was derivatized with phenylisothiocyanate to produce phenylthiocarbamyl alanine and then analyzed by HPLC under the following conditions: column, Wakosil-PTC, 4.0 mm · 250 mm (D) (Wako Pure Chemicals Inc., Japan), solvent A, 60 mM sodium acetate buffer (pH 6.0) in acetonitrile (94:6); and solvent B, 60 mM sodium acetate buffer (pH 6.0) in acetonitrile (40:60).…”
