Induction of Germ Cell-like Cells from Porcine Induced Pluripotent Stem Cells

Abstract: The ability to generate germ cells from pluripotent stem cells (PSCs) is valuable for human regenerative medicine and animal breeding. Germ cell-like cells (GCLCs) have been differentiated from mouse and human PSCs, but not from porcine PSCs, which are considered an ideal model for stem cell applications. Here, we developed a defined culture system for the induction of primordial germ cell-like cells (PGCLCs) from porcine induced PSCs (piPSCs). The identity of the PGCLCs was characterized by observing cell mor… Show more

“…Then, the pPGCs at E15, stage‐equivalent to the E8.5 in the mouse, commence migration towards the genital ridges and, at E18, they start to colonize the genital ridges . Recently, the induction of porcine PGCLCs (pPGCLCs) from porcine iPSCs was reported (Figure ) . Porcine iPSCs first are converted into EpiLCs in the presence of bFGF and activin A for 2 days.…”

“…56 Recently, the induction of porcine PGCLCs (pPGCLCs) from porcine iPSCs was reported ( Figure 4). 57 Porcine iPSCs first are converted into EpiLCs in the presence of bFGF and activin A for 2 days. Then, the disaggregated cultures are kept in the presence of four cytokines (BMP4, LIF, SCF, and EGF) to induce pPGCLCs.…”

“…MSCs do not form teratomas when injected into donor testes and have even been shown to help reestablish the testicular niche following chemical damage (Cakici et al, 2013;Maghen et al, 2017;Vahdati et al, 2017). Male PGCLCs have also been produced from induced pluripotent stem cells (iPSC), which can also be derived from adult animals (Hayashi et al, 2011;Sasaki et al, 2015;Wang et al, 2016a). Further experiments are required to definitively demonstrate that these PGCLCs from different cell types can fulfill the functional definition of a germ cell, i.e.…”

“…The differentiation approach used to generate PGCLCs from human EpiLCs served as template for the evaluation of a similar culture system in pigs using iPSCs. The PGCLC identity was supported by the expression of PGC (BLIMP1, PRDM14, STELLA) and pluripotency markers (OCT4, SOX2), an epigenetic status similar to in vivo PGCs, and transcriptome and gene ontology analyses that revealed a gamete production scheme (Wang et al, ). Also, the recently developed pig EPSCs produced PGCLCs in vitro (Gao et al, ).…”
