Induction of tumouricidal leucocytes by the intranasal application of MTP-PE, a lipophilic muramyl peptide

Abstract: Summary. Single intranasal applications of MTP-PE, a lipophilic muramyl peptide, induce tumouricidal and tumouristatic leucocytes in the lungs of rats. In ex vivo assays the tumouristatic activity was detectable for 8 days after drug administration. By separation of the effector cells on Ficoll-Hypaque gradients, it was shown that both neutrophils and macrophages are responsible for this activity. Using the B16/BL6 melanoma system in mice, there was a high survival rate after repeated intranasal applications o… Show more

“…Given orally to mice, free or liposomal MTP-PE (phosphatidylcholine liposomes) activated both alveolar and peritoneal macrophages, and inhibition of lung and lymph node metastasis was observed [45,46]. Intranasal or intratracheal MTP-PE stimulated macrophages and granulocytes in the lungs of rat, and increased mice survival in the B16 lung metastasis model [47,48]. Intraperitoneal treatment enhanced survival of mice with i.p.…”

“…The commercial preparation of liposomal-MTP-PE used in the clinical trial has been used with success to activate human blood monocytes in vitro [157,161] and murine macrophages both hi vitro and h~ vivo [162,167]. In addition, this liposome-MTP-PE preparation has been found to activate pulmonary macrophages following intranasal administration [191], to produce systemic macrophage activation following oral administration [192] and, as described above, to eliminate spontaneous murine melanoma metastases [177] and autochthonous osteogcnic sarcoma metastases in dogs [183].…”
