Insight into Amyloid Formation Using Congo Red as the Electrochemical Probe

Abstract: The amyloid-b (Ab) aggregation pathway is an important target for the discovery of drugs that can prevent or delay the onset of Alzheimers disease (AD). The electrochemistry of Congo Red (CR) represents a particularly promising tool for screening of Ab-binding therapeutics in a rapid and cost-effective format. The results of the differential pulse voltammetry (DPV) measurements were confirmed using simultaneous UV-vis analysis of the same incubated Ab samples. The early changes in the electrochemical signals w… Show more

“…26 Congo Red was also grafted to a screen-printed carbon electrode via an electrooxidation process to provide electrochemical sensors of the aggregation of amyloid-β oligomers in relation to the onset of Alzheimer's disease. 27 Herein, we wish to explore the propensity of isolated diazotized dyes to modify MWCNTs spontaneously. We concentrate our task on tuning the surface chemical composition of MWCNTs using diazotized dyes simply in water and at RT.…”

“…For example, diazotized Azure A was (electro)grafted to sp 2 carbon nanomaterials for gas sensing and electrocatalytic purposes, , whereas Neutral Red diazonium salt was generated and attached to gold electrodes through the GraftFast process for making optodes for the measurement of pH . “…Redox‐active β‐sheet‐intercalating agents have been used to monitor the kinetics of fibril formation 3537. The structural changes associated with the β‐sheet formation and aggregation result in quantifiable changes in the electrochemical signal of the intercalated compounds.…”

“…Kiessling and co-workers, by immobilizing Ab(1À42) onto an sensor chip, used surface plasmon resonance to screen the various Ab segments for b-sheet breaking peptides [29]. Recently, electrochemical meth-ods have found applications in the studies of Ab aggregation and the dynamic interactions between Ab and a compound of interest [30][31][32][33]. Vestergaard et al were the first to investigate in real time aggregation of Ab peptides by following the variation of the oxidation currents of the tyrosine residue at position 10 (Tyr-10) during the Ab aggregation/fibrillation process [34].…”
