Integrating Fluorinated Polymer and Manganese‐Layered Double Hydroxide Nanoparticles as pH‐activated 19F MRI Agents for Specific and Sensitive Detection of Breast Cancer

Abstract: 19F magnetic resonance imaging (19F MRI) agents capable of being activated upon interactions with cancer triggers are attracting increasing attention, although challenges still remain for precise and specific detection of cancer tissues. In this study, a novel hybrid 19F MRI agent for pH‐sensitive detection of breast cancer tissues is reported, a composite system designed by conjugating a perfluoropolyether onto the surface of manganese‐incorporated layered double hydroxide (Mn‐LDH@PFPE) nanoparticles. The 19F… Show more

“…Then, the fluorescence intensity decreased gradually until 48 h, mostly attributed to the biodegradation (metabolism) of LDH NPs in the acidic TME (pH 6.2–6.8), [ 34 ] consistent with the previous reports. [ 35 ] The blood clearance of LDH NPs from TME may occur to a less extent as the drainage system is not well developed in TME. [ 36 ] Similarly, the ex vivo fluorescence images of the tumors show the strongest ICG signal at 24 h postinjection (Figure 5B,D).…”

“…Previous researches have evidenced that -Mn-O- bond is sensitive to acidic and reductive environments, which is the representative characteristics of tumor microenvironment 22 , 40 . As presented in Figure 3 A, GSH can reduce the -Mn-O- bonds in MnMSN to Mn 2+ , meanwhile GSH was oxidized to GSSG.…”

“…169,170 Zhang et al developed a novel pH responsive 19 F probes, Mn-LDH@PFPE NP, which is activated specifically within the acidic tumor environment. 171 The 19 F MRI signals from NPs are at physiological pH 7.4, but activated at extracellular pH 6.5. In vivo experiments reveal that an intense 19 F MR signal can be detected in the tumor after injection of NPs.…”
