Interaction of Di-2-pyridylketone 2-pyridine Carboxylic Acid Hydrazone and Its Copper Complex with BSA: Effect on Antitumor Activity as Revealed by Spectroscopic Studies

Abstract: Abstract:The drug, di-2-pyridylketone-2-pyridine carboxylic acid hydrazone (DPPCAH) and its copper complex (DPPCAH-Cu) exhibit significant antitumor activity. However, the mechanism of their pharmacological interaction with the biological molecule bovine serum albumin (BSA) remains poorly understood. The present study elucidates the interactions between the drug and BSA through MTT assays, spectroscopic methods and molecular docking analysis. Our results indicate that BSA could attenuate effect on the cytotoxi… Show more

“…The values of Δ H , Δ S , and Δ G for Dp44mT binding to HSA are listed in Table 4 . Based on the view of Ross and Subramanian, negative Δ H and Δ S values implied that the interaction between Dp44mT and HSA occurred via hydrogen bonding and van der Waals interactions, consistent with a study of a Dp44mT analog [ 16 ]. The negative value of Δ G also revealed that the interaction process was spontaneous.…”

“…The synchronous fluorescence spectra of 1–6 showed hypochromism with negligible shift in the emission, whereas the tryptophan and tyrosine residue of the serum albumins showed hypochromism accompanied with 2–3 nm of red shift (Figure , and Figure S8‐S11). This red shift indicates that that 1–6 effectively bind with serum albumins and affect the conformation of the Trp micro regions: hydrophobicity of Trp residues decreased and polarity increased .…”
