Involvement of human decidual cell-expressed tissue factor in uterine hemostasis and abruption

Abstract: Vascular injury increases access and binding of plasma-derived factor VII to perivascular cell membrane-bound tissue factor (TF). The resulting TF/VIIa complex promotes hemostasis by cleaving pro-thrombin to thrombin leading to the fibrin clot. In human pregnancy, decidual cellexpressed TF prevents decidual hemorrhage (abruption). During placentation, trophoblasts remodel decidual spiral arteries into high conductance vessels. Shallow trophoblast invasion impedes decidual vascular conversion, producing an inad… Show more

“…Thrombin also promotes membrane rupture by activating MMPs or protease-activated receptors in the amnion (64)(65)(66). Activation of the coagulation cascade that releases thrombin and other proteases may be initiated by decidual tissue factor, which can be released by infection, inflammation, hypoxia, or oxidative stress (67). In addition to thrombin, other proteases, such as elastase, may be involved in contraction signals, releasing fibronectin and promoting the breakdown of the chorion-decidual fusion (68).…”

“…Implantation of the human blastocyst involves trophoblastic invasion deep into the decidual cells of the uterus. Shallow invasion leads to inadequate decidual vascular formation and subsequent poor uteroplacental blood flow (Lockwood, et al, 2009). Placental ischaemia occurs and is implicated in the pathogenesis of placental abruption and of preterm birth (Vintzileos, 2008, Ananth, et al, 2007.…”
