Abstract: A total of 143 samples of meat were examined in this study, theserepresents 67 samples of locally minced meat, 31 samples of importedminced meat and 45 samples of imported chicken meat. This study was aimed to isolate a locally higher cytosine deaminase producer Escherichia coli. Ten E. coli bacterial isolates were obtained from twenty-five urine samples collected from Al-Imamein AlKadhumain medical city hospital in Baghdad. These isolates were identified by cultural, microscopical and biochemical tests, identifications also done by vitek 2 system. Cytosine deaminase activity was assayed for these isolates and it was found that all isolates were enzyme producer with variable activities and E. coli E9 was the most efficient with specific activity of (0.350 U/mg).

Escherichia coli O157:H7 is an important food borne pathogen has been linked to foods of bovine origin and fresh produce. Rapid and sensitive identification of this dangerous pathogen is important for patient management and for prompt epidemiological investigations.This study aimed to detect Escherichia coli O157:H7 presence in samples from diarrhea patients, and food (imported beef and lettuce) and characterize by selective media, biochemical tests, latex agglutination test, and compared with PCR technique. during the period from September 2012 to February 2013 A total 120 samples were collected 50 from diarrhea patients and 70 food samples including (45 imported beef and 25 lettuce) All samples were screened to detect the presence of non-sorbitol fermenting colonies (NSF) on sorbitol Mac Conkey agar supplemented with Cefixime (C-SMAC). A total of 120 isolates, 22 (44%) from diarrhea patients, 18 (40%) from meat and 9 (36%) from lettuce were non-sorbitol fermenting, E. coli isolates were serotyped as E. coli O157:H7 by latex agglutination test, 10(20%) isolates of diarrhea patients, 7 (15.5%) isolates of meat and 5 (20%) isolates of lettuce, Results of the genetic diagnosis using polymerase chain reaction (PCR)with specific primers (eaeA and rfbE genes) by MPCR revealed that 10(20%) isolates of diarrhea patients5 (11.1%) isolate from meat and 5(20 %) of lettuce samples were positive.PCR has become a very rapid and reliable tool for the molecular diagnosis of E. coli O157.

Investigation of Infectious Laryngotracheitis Virus in Broiler Flocks in Sulaymaniyah Province, Iraq

Infectious laryngotracheitis (ILT) is an acute contagious upper respiratory tract infection of ‎chickens and other birds, caused by Gallid herpesvirus1 (GaHV-1), which has economic ‎importance in the poultry industry. There was no scientific data about the incidence of the ‎disease in broiler farms in Sulaymaniyah province/Iraq. Therefore, this study aimed to ‎investigate ILTV infection in broiler farms in that region. Clinically infected birds from 89 ‎broiler flocks that had respiratory distress, coughing, gasping, tracheal rales, nasal ocular ‎discharge, and congested trachea with purulent exudate, hemorrhagic tracheitis with/or without ‎necrotic changes were investigated. The DNA was extracted from the pooled samples, ‎including tracheal secretion, trachea, and lung tissue. Primers specific to the thymidine kinase ‎gene (tdk) of ILTV- were used in PCR to detect the virus. A phylogenic tree was generated to ‎track the virus’s origin. The study revealed that the rate of infection with ILTV among broiler ‎farms was 2.2% (2/89) in the region. The sequencing analysis showed that the ILTV isolated in ‎the area was closely related to the reported strains in the United States and Brazil (MN643591.1 ‎and S83714.1); and had a sequence identity of 98.27% to the taxon ‎JQ217378.1. In conclusion, the study reported that one of the causes of the respiratory viral ‎infection in broiler flocks even at younger ages was related to ILTV. Partially sequenced tdk ‎gene of the virus showed that the circulated serovar in the region had some nucleotides and ‎amino acids differences with the worldwide reported serovars. This should be taken into ‎consideration in the poultry industry by doing further investigation‎.
