Isolation of recombinant strains with enhanced pectinase production by protoplast fusion between Penicillium expansum and Penicillium griseoroseum

Abstract: Protoplast fusion between complementary auxotrophic and morphological mutant strains of Penicillium griseoroseum and P. expansum was induced by polyethylene glycol and calcium ions (Ca 2+ ). Fusant strains were obtained in minimal medium and a prototrophic strain, possibly diploid, was chosen for haplodization with the fungicide benomyl. Different recombinant strains were isolated and characterized for occurrence of auxotrophic mutations and pectinolytic enzyme production. The fusant prototrophic did not … Show more

“…Different methodologies which have been used to achieve these goals include protoplast production and regeneration, mutant isolation and characterization, protoplast transformation and isolation and characterization of genes coding pectinolytic enzyme in P. griseoroseum and P. expansum. (5,9,12,13,16,21,33,(34)(35)(36)38,41). Incompatibility was observed between these two species for the formation of heterokaryons and diploids when conidia were mixed (41).…”

“…(5,9,12,13,16,21,33,(34)(35)(36)38,41). Incompatibility was observed between these two species for the formation of heterokaryons and diploids when conidia were mixed (41). When the parasexual cycle of P. expansum was analyzed, it was noted that diploids were rather unstable and that recombinant sectors could easily be isolated (21).…”

“…It is also higher than the increase observed for others enzymes evaluated in DG-resistant mutants for other fungal species 20, 33. A 1.2-fold improvement in PL production has also been obtained by protoplast fusion between P. griseoroseum and P. expansum 12 . Future attempts could evaluate the PL production after protoplast fusion between mutant M03 and other strain showing good PL production or between mutants M03 and M04.…”
