“…The K,,, values for L-tyrosine ranged from -100 to -250,uM, those for B-MPH4 being from -180 to -250 PM. The K, values for L-tyrosine are similar to those obtained from the purified rat [20] and bovine [21] enzymes. The low-affinity form of TH to pteridine (Km -200 PM) reportedly changes to the high-affinity form (K,,, -2OpM) through protein phosphorylation [22,23].…”

“…1 7 Since tyrosine hydroxylase, the rate-limiting enzyme for DA synthesis, requires molecular oxygen, a complete ischemia would reduce the synthesis of DA. The high affinity of tyrosine hydroxylase for oxygen, 18 however, may leave the brain aloof from the lack of the substrate, molecular oxygen. In addition, the increase in cyclic AMP observed in ischemic brain 19 -M would activate tyrosine hydroxylase through cAMP-dependent protein-kinase.…”

“…4 BH 4 ) is a natural cofactor for tyrosine hydroxylase (1,2), a rate-limiting enzyme for biosynthesis of catecholamines such as dopamine (DA), noradrenaline and adrenaline, and also a cofactor for tryptophan hydroxylase (3), a rate-limiting enzyme for biosynthesis of 5-hydrox~tr\'ptamine \.;;-HT). Because the concentration of 6R-BH 4 in the brain (4-7) is low compared with the Michaelis constants (Km values) of tyrosine hydroxylase (8)(9)(10)(11)(12)(13) and tryptophan hydroxylase (9,14) for 6R-BH 4 , tissue levels of 6R-BH 4 are considered to regulate the activities in vivo of thess enzymes. Our previous findings that intracerebroventricular administration of 6R-BH4 resulted in enhanced hydroxylation of tyrosine and tryptophan in the rat brain (15) supported this possibility.…”
