Latent ECSO VIRUS Infections of Sucking Pigs: I. Isolation of latent enteroviruses from tissue cultures, obtained from kidneys of apparently healthy sucking pigs

Abstract

Summary Primary tissue cultures were prepared from kidneys of 52 clinically healthy sucking pigs. The kidney tissues yielded trypsinized cells with preserved dividing ability and metabolism. As a result of their multiplication, a confluent cell sheet was formed on the glass surface in the course of 72 to 96 hours incubation at 37° C. After this period eleven of them showed spontaneous cytopathogenic changes and viruses were isolated which were resistant to ethyl ether, chloroform, trypsin, sodium desoxycholate and saponin. Ten of the 11 viruses were tested and were neutralized with hyperimmune rabbit sera obtained against standard porcine enteroviruses. In the blood of the pig virus carriers a high level titer of neutralization antibodies against homologous enteroviruses was established. The results of these investigations suggest that a latent enterovirus infection of the pig kidney cells was present in vivo. The role is discussed of an “antibody‐enterovirus” complex as a form of existence of the virus particles in the cell‐virus system, its disintegration in the nutrient tissue culture medium and the subsequent release of unmasked, virulent enterovirus particles, able to infect susceptible cells of the tissue cultures. Zusammenfassung Latente ECSO‐Virus‐Infektionen bei Saugferkeln I. Isolierung latenter Enteroviren aus Nierengewebekulturen von gesunderscheinenden Saugferkeln Aus den Nieren von 52 klinisch gesunden Saugferkeln wurden Gewebe‐Primärkulturen hergestellt. Aus dem Nierengewebe wurden trypsinierte Zellen gewonnen, bei denen Teilungsfähigkeit und Stoffwechsel erhalten waren. Nach 72‐ bis 96stündiger Bebrütung bei 37° C bildete sich auf der Glasoberfläche ein konfluierender Zellbelag aus. Danach entwickelten sich in 11 Kulturen spontan zytopathogene Veränderungen, und es wurden Viren isoliert, die gegenüber Äthyläther, Chloroform, Trypsin, Na‐desoxycholat und Sapronin resistent waren. Von ihnen wurden 10 getestet und mit einem Kaninchen‐Hyperimmunserum gegen Standard‐Enteroviren vom Schwein neutralisiert. Im Blut der Virusträger fand sich ein hoher Titer neutralisierender Antikörper gegen die homologen Enteroviren. Diese Befunde machen eine in vivo bestehende Enterovirus‐Infektion der Schweinenierenzellen wahrscheinlich. Die Bedeutung eines “Antikörper‐Enterovirus”‐Komplexes als einer Existenzform von Viruspartikeln im Zell‐Virus‐System wird diskutiert im Hinblick auf den Zerfall im Gewebekultur‐Nährmedium und die folgende Freisetzung unmaskierter virulenter Enteroviruspartikel, die fähig sind, empfängliche Zellen der Gewebekultur zu infizieren.
