Low‐Frequency Ultrasound Sensitive Piezo1 Channels Regulate Keloid‐Related Characteristics of Fibroblasts

Zixi Jiang,
Ziyan Chen,
Yantao Xu
et al.

Abstract: Keloids are benign fibroproliferative tumors that severely diminish the quality of life due to discomfort, dysfunction, and disfigurement. Recently, ultrasound technology as a noninvasive adjuvant therapy is developed to optimize treatment protocols. However, the biophysical mechanisms have not yet been fully elucidated. Here, it is proposed that piezo‐type mechanosensitive ion channel component 1 (Piezo1) plays an important role in low‐frequency sonophoresis (LFS) induced mechanical transduction pathways that… Show more

“…Although rodent models cannot fully recapitulate the development of keloids, pharmacological inhibition or knockdown of YAP/TAZ or PIEZO1 in keloid fibroblasts attenuated a series of pro-fibrotic phenotypes in vitro , including fibroblasts’ proliferation, contraction, migration, and collagen synthesis. Moreover, elevated protein levels of YAP/TAZ and PIEZO1 have been observed in clinical specimens of keloids compared to normal human skin (16, 17). Together, these results suggest a contribution of mechano-transduction in fibroblasts to keloid pathogenesis.…”
