Long noncoding RNA THAP9-AS1 is induced by Helicobacter pylori and promotes cell growth and migration of gastric cancer

Abstract: Background: Long noncoding RNAs (LncRNAs) have been confirmed to play crucial roles in cancer biology. Gastric cancer (GC) is the third leading cause of cancer related death, and Helicobacter pylori (H. pylori) is the major risk factor for GC. In this study, we focused on the roles of H. pylori-related lncRNAs in the progression of GC. Method: Differentially expressed lncRNAs were identified through RNA-seq analysis of H. pylori-infected GC cells. Results: We found that the expression of the lncRNA THAP9-AS1 w… Show more

“…THAP9-AS1 is a recently annotated lncRNA and has been shown to be upregulated in pancreatic cancer and neutrophil apoptosis and downregulated in case of septic shock (35)(36)(37) . In this study, we have quantified THAP9 and THAP9-AS1 transcript expression under various stress conditions in the S-phase of cell cycle.…”

“…This leads us to speculate that there exists a stress specific regulation of THAP9 and THAP9-AS1 gene-pair that switches to THAP9-AS1 under stress conditions. This also might explain the upregulation of THAP9-AS1 in cancer cells which are naturally stressed (3537). Moreover, it is possible that both these genes play an unknown role in DNA repair and this might be responsible for their increased expression under certain stress conditions.…”

“…Although the effect of the two lncRNAs in sepsis remains unclear, their roles have been well studied in various human diseases. THAP9-AS1, induced by Helicobacter pylori , can promote cell growth and migration of gastric cancer [ 40 ]. THAP9-AS1 can also promote pancreatic ductal adenocarcinoma growth and lead to a poor clinical outcome via sponging miR-484 and interacting with YAP [ 41 ].…”

“…4 Reportedly, lncRNAs can markedly regulate the progression of multiple cancers including GC. [5][6][7] For instance, as an oncogenic lncRNA, lncRNA AFDN-DT can inhibit GC cell growth through transcriptional regulation; 8 as a tumorsuppressive factor, LINC00460 is significantly upregulated in GC tissues, and promotes the malignant biological behaviors of cancer cells via epigenetically silencing CCNG2. 9 LncRNA C-terminal binding protein 1 antisense RNA 2 (CTBP1-AS2) is not only related to the susceptibility to type 2 diabetes 10 but also closely connected with the recurrence of papillary thyroid cancer.…”
