Mainstream Smoke Chemistry and in Vitro and In Vivo Toxicity of the Reference Cigarettes 3R4F and 2R4F

Abstract: A new reference cigarette, the 3R4F, has been developed to replace the depleting supply of the 2R4F cigarette. The present study was designed to compare mainstream smoke chemistry and toxicity of the two reference cigarettes under the International Organization for Standardization (ISO) machine smoking conditions, and to further compare mainstream smoke chemistry and toxicological activity of the 3R4F cigarette by two different smoking regimens, i.e., the machine smoking conditions specified by ISO and the Hea… Show more

“…In this model, the cytotoxic effect of WS was primarily driven by VP components and on HCI smoking there was a general reduction in the majority of the particulate and VP toxicants when expressed per mg of TPM, tar or nicotine (data not shown), an observation in agreement with published studies (Roemer et al, 2012 there was a trend towards increased IL-8 secretion following HCI exposure. The reason for this is unclear.…”

“…The composition of the reference cigarettes as well other characteristics of this reference tobacco are reported by the supplier [36]. The proximate analysis of this tobacco provides the following results: (8.2% of moisture, 69.9% of volatiles, 10.3% of fixed carbon and 11.9% of ashes.…”

“…Mutagenicity of TPM was assessed in the plate incorporation Salmonella Reverse Mutation Assay (Maron and Ames, 1983) according to the respective OECD guideline 471 (OECD, 1997a) as previously described (Roemer et al, 2012). Determinations were performed in the presence and in the absence of a metabolic activation system.…”
