Manganese Active Transport in Escherichia coli

Abstract: Manganese was accumulated by cells of Escherichia coli by means of an active transport system quite independent of the magnesium transport system. When the radioisotope 54 Mn was used, manganese transport showed saturation kinetics with a K m of 2 × 10 −7 m and a V max of 1 to 4 nmoles/min per 10 12 cells at 25 C. The manganese trans… Show more

“…Interestingly, a related Mn(II) transport system was described in B. subtilis (Bhattacharyya, 1975) and was shown to constitute the primary uptake pathway of Cd(II) ions (for recent reviews, see Silver and Walderhaug, 1992). Some evidence suggests that MntH is in fact the Mn(II) transporter previously described in E. coli (Bhattacharyya, 1970;Silver et al 1970). First, E. coli MntH mediates high-capacity manganese ion uptake (Fig.…”

“…We have studied the effect of a yfeP null mutation and of overexpression of the corresponding protein on bacterial divalent cation transport. The functional characteristics presented here suggest that it encodes the previously described proton-dependent manganese transport system of E. coli (Bhattacharyya, 1970;Silver et al, 1970), here designated MntH.…”

“…Manganese(II) uptake and efflux and iron(II) uptake were determined by incubating cells in DT medium (0.4% tryptone peptone, 0.25% NaCl) with 54 Mn 21 (Amersham) as used by Silver et al (1970), with triplicate samples at each time point or concentration. Cells were then processed essentially as for Mg 21 transport assays from this laboratory (Grubbs et al, 1989).…”

“…Mil2+ is a trace element generally required by 0,-evolving photosynthetic organisms ( MARTIN 1966, 1969). A few bacteria lacking 0, formation have also been found to show Mn2+ requirement for growth (SILVER and JASPER 1977). However, it has been difficult t o demonstrate a Mn2+ requirement for growth of Escherichiu coli (SILVER et a2.…”
