Morphology plays a vital role in determining the biological effects of silica nanoparticles (NPs), but its influence on the toxicity of silica NPs in endothelial cells (ECs) is still inconclusive. We synthesized five kinds of Santa Barbara 15 amorphous (SBA‐15) particles with different shapes and added them to human umbilical vein endothelial cells (HUVEC). After 24 After incubation and treatment with 100 ml, more than 80% of the cells are still alive. The microgram/ml of SBA‐15 indicates that SBA‐15 has high biocompatibility. Fibrous SBA‐15 (5) leads to the highest Si element concentration in HUVEC. No NP reduces the release of NO, and NO is an important signaling molecule in the vascular system. Only the aggregated spherical SBA‐15 (3) will moderately reduce the endothelial nitric oxide synthase (eNOS) protein. Regarding transcription factors regulating eNOS, we found that all SBA‐15 types significantly increased Kruppel‐like factor 2 (KLF2) protein, irregular SBA‐15 (1), non‐aggregated spherical SBA‐15 (2) and aggregation The spherical SBA‐15 (3) greatly reduces KLF4 by more than 50%. Overall, our results indicate that SBA‐15 with different morphologies can be internalized into HUVEC and only cause moderate cytotoxicity. All silica NPs have the smallest effect on the NO‐eNOS pathway, but the irregular spherical SBA‐15 reduces the eNOS modifier KLF4. The rod‐shaped SBA‐15 (4) seems to have higher biocompatibility because they are internalized and have negligible adverse effects on HUVEC. These results provide new evidence for the toxic effects of different forms of silica nanoparticles on HUVEC.

Nanozyme‐based chemodynamic therapy (CDT) has emerged as an effective cancer treatment because of its low side effects and without the requirement of exogenous energy. The therapeutic effect of CDT highlights the pivotal importance of active sites, H2O2 supplement and the glutathione (GSH) depletion of a nanozyme. The construction of a single kind of catalyst with multiple functions for the enhanced CDT is still a big challenge. In this work, seven types of bimetallic nanoparticles are synthesized using a metal–organic framework (MOF) as a stable host instead of a Fenton or Fenton‐like ions supplier. Among them, Cu‐Pd@MIL‐101 with an alloy loading of 9.5 wt% modified by PEG (9.5% CPMP) is found to exhibit the highest peroxidase (POD) like activity combined with a superoxide dismutase (SOD) mimic activity and the function of GSH depletion. The in vivo results suggest that the stable and ultrafine nanoparticles possess favorable CDT effect for tumor and good biosafety as well as biocompatibility. This work has provided a credible strategy to construct nanozymes with an excellent activity and may pave a new way for the design of enhanced tumor CDT treatment.

Novel metal and metal oxide‐based nanocomplexes are being developed due to their superior properties compared with nanoparticles (NPs) based on single composition. In this study, we synthesized Ag‐coated ZnO (Ag/ZnO) and Ag‐doped ZnO (Ag@ZnO) NPs. The cytotoxicity and mechanisms associated with the synthesized NPs were investigated to understand the influence of Ag positions on biocompatibility of the NPs. After exposure to human umbilical vein endothelial cells (HUVECs), Ag/ZnO, Ag@ZnO, and ZnO NPs all significantly induced cytotoxicity, but the cytotoxic effects of Ag/ZnO and Ag@ZnO NPs were more modest in comparison with ZnO NPs. At cytotoxic concentrations, all NPs significantly induced intracellular Zn ions, which suggested a role of excessive Zn ions on cytotoxicity of NPs. All types of NPs significantly induced the expression of endoplasmic reticulum (ER) stress genes including DNA damage‐inducible transcript 3 (DDIT3), X‐box binding protein 1 (XBP‐1), and ER to nucleus signaling 1 (ERN1), but Ag/ZnO and Ag@ZnO NPs were less effective to induce DDIT3 and XBP‐1 expression compared with ZnO NPs. Not surprisingly, only ZnO NPs significantly induced the expression of caspase 3. Combined, the results from this study showed that Ag/ZnO and Ag@ZnO NPs were less cytotoxic and less potent to induce ER stress gene expression compared with ZnO NPs, but there were no significant differences between Ag/ZnO and Ag@ZnO NPs. Our results may provide novel understanding about the biocompatibility of Ag–ZnO nanocomplexes.
