Meal Anticipatory Rise in Acylated Ghrelin at Dark Onset is Blunted After Long-Term Fasting in Rats

Abstract: Ghrelin is a 28 amino acid acylated peptide originally characterised for its capacity to stimulate growth hormone secretion. Ghrelin is also an orexigenic and adipogenic hormone and is thought to be a signal to increase locomotor activity in anticipation of a scheduled meal. Although ghrelin is considered to be up-regulated during fasting, there are still conflicting data regarding the impact of starvation on ghrelin secretion. To test whether the secretory pattern of acylated ghrelin is altered during fasting… Show more

“…Corticosterone in this group was elevated at ZT0 and ZT4 compared to the ZT16 group, and slightly decreased at ZT12. This pattern conforms to previous reports that in daytime fed rats, corticosterone rises in anticipation of the daytime meal, but also shows a nocturnal peak, reflecting a continued influence of the LD entrained SCN pacemaker [36] .…”

“…Furthermore, using electrophysiological recording, we have shown that ghrelin‐induced activation of NPY neurones is abolished in GHS‐R null mice . Interestingly, ghrelin has been demonstrated to exhibit daily rhythms of secretion that are synchronised by meal timing in both humans and rodents, showing pre‐prandial increases in circulating acyl‐ghrelin levels in anticipation of scheduled meals . Furthermore, transcriptomic studies have identified GHS‐R as one of the top G‐protein coupled receptors to be significantly up‐regulated by food deprivation compared to fed state in isolated AgRP neurones .…”

“…Ghrelin is perhaps best known as a circulating hunger signal necessary for meal initiation and meal anticipation with a secretion occurring in a pulsatile manner starting with a preprandial rise and postprandial fall 1 h after food intake (Ariyasu et al, 2001; Cummings et al, 2001; Tschöp et al, 2001; Zizzari et al, 2011; Merkestein et al, 2012). Moreover, whatever the route of injection, ghrelin increases food intake both in humans and animals (Wren et al, 2001a,b).…”
