Measurement comparability of insulin assays using conventional immunoassay kits

Abstract: Immunoassays are analytical methods that achieve the detection and quantification of analytes, particularly peptides and proteins in biological samples, through the formation of a stable complex between the analyte and a specific antibody. They represent very selective and sensitive techniques that have found application in several areas such as clinical chemistry, bioanalysis, pharmaceutical analysis, toxicological analysis, and environmental analysis. [1][2][3] Owing to their capacity for high throughput and… Show more

“…Notably, the ability to measure glucose in the interstitial space using an enzymatic method, which does not require multiple washes, allows for the continuous measurement of glucose in these devices. Unfortunately, measuring insulin requires using either ELISA or radioimmune assays 64 , thereby currently making immediate readings impossible. Nevertheless, even in the absence of continuous insulin monitors, it may be possible to monitor postprandial glucose levels within the frameworks of Kraft patterns.…”

“…The isolated β-cells of the pancreas were incubated in Krebs Ringer bicarbonate buffer supplemented with different concentrations of glucose solution (3.3 mM, 8.3 mM, 11.1 mM, and 16.7 mM) for approximately 30 min [ 44 ]. After incubation for 30 min, the supernatant was collected for insulin secretion assay (Invitrogen; Massachusetts, USA; Cat #BMS2003) as per the method of [ 45 ]. To quantify the ATP levels within the pancreas, pancreatic β cells underwent lysis, and the ATP content was assessed using an ATP assay kit (Thermofisher; Massachusetts, USA; Cat #A22066) as per method of [ 46 ].…”

“…For insulin, it was beneficial that the samples had been examined by a clinically approved ELISA kit prior to being analyzed by the LC-MS method [30], as a suitable calibration concentration range could easily be selected. For the other hormones, somatostatin-14 and glucagon, a gold standard for the determination of the concentrations has yet not been established [31][32][33].…”
