Melatonin promotes sorafenib‐induced apoptosis through synergistic activation of JNK/c‐jun pathway in human hepatocellular carcinoma

Abstract: Melatonin has been shown to exert anticancer activity on hepatocellular carcinoma (HCC) through its antiproliferative and pro-apoptotic effect in both experimental and clinical studies, and sorafenib is the only approved drug for the systemic treatment of HCC. Thus, this study was designed to investigate the combined effect of melatonin and sorafenib on proliferation, apoptosis, and its possible mechanism in human HCC. Here, we found that both melatonin and sorafenib resulted in a dose-dependent growth inhibit… Show more

“…However, no study has investigated the contribution of Mst1 to renal I/R injury. Moreover, Mst1 has been reported to be associated with mitochondrial damage in myocardial ischaemia reperfusion injury [27], pancreatic cancer cells [28], and spinal cord trauma [29]. This evidence supports the possibility that Mst1 can be considered the upstream regulator of mitochondrial homeostasis during acute cell stress.…”

“…sorafenib in hepatocellular carcinoma [56], cisplatin in hepatocellular carcinoma [57], fluorouracil in oesophageal squamous cell carcinoma [58], etc. Although diverse mechanisms for melatonin-induced apoptosis have been suggested, many of them have the same denominator — ER stress.…”

“…Total RNA was extracted from the cells using TRIzol® reagent (Invitrogen Life Technologies, Carlsbad, CA, USA) and was reverse-transcribed into a total of 1 µl (60 ng/µl) cDNA using a One-Step RT-PCR kit (TransGen Biotech Co., Ltd., Beijing, China), according to the manufacturer’s instructions [35]. Quantification of gene expression was performed using an ABI PRISM 7500 Sequence Detection system (Applied Biosystems Life Technologies, Foster City, CA) with SYBR® Green (TransGen Biotech Co., Ltd.) as previously described [33].…”
