Metabolic engineering of β-oxidation in Penicillium chrysogenum for improved semi-synthetic cephalosporin biosynthesis

Abstract: Industrial production of semi-synthetic cephalosporins by Penicillium chrysogenum requires supplementation of the growth media with the side-chain precursor adipic acid. In glucose-limited chemostat cultures of P. chrysogenum, up to 88% of the consumed adipic acid was not recovered in cephalosporin-related products, but used as an additional carbon and energy source for growth. This low efficiency of side-chain precursor incorporation provides an economic incentive for studying and engineering the metabolism o… Show more

“…Penicillium chrysogenum has had a key historical role in the development of industrial microbiology, and is currently one of the most important species in the biotechnological industry as producer of penicillin [ 5 ] and other β-lactam antibiotic precursors [ 6 ]. The P. chrysogenum heterotrimeric Gα protein Pga1, belonging to the fungal subgroup I, has been characterized and shown to participate in the regulation of the global developmental program of the fungus, from spore germination to conidia formation [ 7 9 ].…”

“…through metabolic network modelling, for the production of enzymes or secondary metabolites. Metabolic engineering strategies have previously been used with great success to redesign metabolic fluxes for secondary metabolite production in filamentous fungi 35 36 37 and for heterologous expression 38 39 40 . Enabling the transfer of genes or gene clusters to well-known expression hosts serves as an important step in the development of efficient cell factories that can be used for economically viable production of secondary metabolites in the food and pharmaceutical industry 41 .…”

“…The filter containing the mycelium was immediately wrapped in aluminum foil, quenched in liquid nitrogen and stored at -80°C. Samples were processed as described [5,6,8,10]. Acquisition and quantification of microarray images and data filtering were performed using Affymetrix GeneChip Operating Software (GCOS version 1.2).…”
