Methodological Guidelines to Study Extracellular Vesicles

Abstract: Owing to the relationship between extracellular vesicles (EVs) and physiological and pathological conditions, the interest in EVs is exponentially growing. EVs hold high hopes for novel diagnostic and translational discoveries. This review provides an expert-based update of recent advances in the methods to study EVs and summarizes currently accepted considerations and recommendations from sample collection to isolation, detection, and characterization of EVs. Common misconceptions and methodological pitfalls … Show more

“…Blood samples for MV-PGC measurement of septic shock (SS) patients and age-and sex-matched healthy controls were collected and processed according to the current International Society on Thrombosis and Haemostasis guidelines [28,29]. Briefly, after a light tourniquet was applied, samples were drawn from the antecubital vein using a butterfly device with a 21-gauge needle.…”

“…Thirty-four consecutive adult patients (mean age: 62 years; range: 19–93 years) with a diagnosis of SS according to the guidelines of the International Sepsis Definition Conference [28] were prospectively enrolled after admission in an intensive care unit. Written informed consent was obtained from the patients or their relatives, and the study was approved by a local medical ethics committee (CPP Sud Méditerranée N° 13–67) according to the Declaration of Helsinki.…”

“…This assay outperformed current predictive models pancreatic cancer patients ( n  = 100). During the study, however, knowledge of blood handling/collection has evolved and the FGT test is amenable to great improvements [18]. Taken together, given the fact that each year about 10,000,000 new cases of VTE are described, of which 20% are associated with cancer, further exploring the coagulant properties of EVs to predict VTE in certain types of cancer, is promising.…”

“…UF uses a membrane filter with pressure, either directly applied or generated by a centrifuge/ultracentrifuge [65]. UF can rapidly isolate EVs from body fluids [66,67], but the yield is low because of the loss of small size EVs, which have a similar size to protein aggregates, necessary to guarantee the purity [68].…”
