Micronucleus assay in bovine lymphocytes after exposure to bisphenol A in vitro

Abstract: Bisphenol A (BPA) [2,2-bis-(4-hydroxyphenyl) propane] is an important industrial agent, made by combining acetone and phenol, that is used extensively as a monomer in the production of polycarbonate plastics and as a precursor of epoxy resins. Micronucleus assays have served as an index of cytogenetic damage in in vivo and in vitro studies. We studied the genotoxic and cytotoxic effects of BPA on bovine peripheral lymphocytes in vitro. Lymphocyte cultures from two donors were exposed to four different concentr… Show more

“…The genotoxicity of BP-A has been evaluated in several in vitro and in vivo studies, although with controversial results. [22][23][24]26 Here, we discuss data obtained from our in vitro study conducted in order to establish the clastogenic and aneugenic potential of BP-A on human cultured lymphocytes. The results obtained in the present in vitro study with CAs and MNs assays evidenced a possible clastogenic effect of the BP-A on human lymphocytes, also at the concentration of 0.05 mg/mL that represents the RD established for humans by the US EPA.…”

“…In one of these (Santovito et al, 2018 [RefID 11220]), MN frequency was also measured. A study of MN in bovine peripheral blood lymphocytes also reported positive findings (Šutiaková et al, 2014 [RefID 7026]).…”

“…In a previous study, lymphocytes were isolated from the peripheral blood of bovines, and the genotoxic effect of BPA was determined at different concentrations, 1 Â 10 À4 , 1 Â 10 À5 , 1 Â 10 À6 , and 1 Â 10 À7 mol/L. And it was found that nuclear divisions for cell proliferation did not change with BPA application, but the number of micronuclei was enhanced with increasing concentrations of BPA (Sutiaková et al, 2014). In our study, we did not examine cell proliferation, although as a result of BPA exposure, the number of CD3 positive cells was decreased in both groups of day 21 and 45.…”
